Serum from ten individual healthy human female donors was measured in duplicate.

Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MASP2 concentration was determined to be 343.24 ng/mL with a range of 56.31 – 528.11 ng/mL.

Interpolated concentrations of native MASP2 in human serum and plasma samples.

The concentrations of MASP2 were measured in duplicates, interpolated from the MASP2 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1%, plasma (citrate) 1%, plasma (heparin) 1% and plasma (EDTA) 1%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MASP2 concentration was determined to be 484.87 ng/mL in serum, 561.89 ng/mL in plasma (citrate), 500.49 ng/mL in plasma (heparin), and 404.43 ng/mL in plasma (EDTA).

How the assay works

Human MASP2 SimpleStep ELISA^{^®}employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA^{^®} plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. MASP2, Mannan-binding lectin serine protease 2, plays a key role in the complement activation cascade. This protein is part of a larger mannose-binding complex which senses foreign material, activates C3 convertase, and perpetuates the immune response. The immunogen from which the antibodies in this kit were raised represents the N-terminal half of the protein.

MASP-2 also known as Mannan-binding lectin serine protease 2 is an important component of the innate immune system. This protein has a molecular mass of approximately 76 kDa and is expressed in the liver before being released into the bloodstream. MASP-2 primarily acts during the lectin pathway of the complement system where it plays an essential role in immune defense. The protein activates the complement cascade upon binding with carbohydrate molecules on pathogen surfaces contributing to opsonization and direct lysis of pathogens.

Biological function summary

MASP-2 associates with MBL (Mannose-binding lectin) and forms a complex critical for the lectin pathway's function. In this complex MASP-2 initiates the cleavage of complement proteins C4 and C2 forming the C3 convertase. This action enhances the immune response by promoting the clearance of pathogens. MASP-1 is another protease that works closely with MASP-2 in this pathway although the two have distinct roles—MASP-2 is primarily responsible for the activation of the downstream components of the cascade.

Pathways

MASP-2 plays an integral role in the complement system and particularly in the lectin pathway which is one of the three pathways of complement activation. It connects with both the classical and alternative pathways through the formation of C3 convertase a critical step in the complement cascade. MASP-2 is connected to conditions like autoimmune diseases and recurrent infections due to its involvement in the complement system. For instance MASP-2 deficiency can lead to increased susceptibility to infections reflecting its role in pathogen clearance. The protein has been studied in relation to systemic lupus erythematosus where dysregulation of the complement pathway can exacerbate disease symptoms. Proteins such as MBL and MASP-1 which interact closely with MASP-2 also have roles in these disease states and further illustrate the importance of proper complement system functioning in disease prevention and control.

Precursor of a serum protease that activates the lectin pathway of the complement system, a cascade of proteins that leads to phagocytosis and breakdown of pathogens and signaling that strengthens the adaptive immune system (PubMed : 11527969, PubMed : 22691502). The lectin complement system is activated following association of lectins, such as MBL2, FCN1, FCN2 or FCN3, to carbohydrates on the pathogen surface (PubMed : 22691502, PubMed : 22966085). MASP2 is cleaved and activated by MASP1 in response to lectin-binding to pathogen carbohydrates (PubMed : 10946292, PubMed : 22949645, PubMed : 22966085, PubMed : 9087411). Can activate prothrombin to thrombin (PubMed : 39924859).. Mannan-binding lectin serine protease 2 B chain. Serine protease component of the lectin complement pathway, which catalyzes cleavage and activation of C2 and C4, the next components of the complement pathway.
