Human IgG ELISA kit is a single-wash, 90-minute SimpleStep ELISA® assay designed for the quantification of human Immunoglobulin G (IgG) with a sensitivity of 0.02 ng/mL. The assay uses a simple Mix-Wash-Read protocol with just one incubation and wash step.

IgG levels in individual healthy donors.

Ten individual healthy donors were evaluated for the presence of IgG in serum using this assay. Results were interpolated from the standard curve in Sample Diluent NS and corrected for sample dilution (1 : 5x106). The mean level of IgG was found at 12.5 mg/mL with a range of 9 – 18.4mg/mL.

Human IgG ELISA Kit ab195215 is a rapid single-wash 90-min ELISA kit to measure Human IgG in tissue lysate, cerebral spinal fluid, saliva, urine, cell culture supernatant, milk, serum and plasma. This SimpleStep ELISA® sensitivity is 0.02 ng/mL.

How the assay works

Human IgG SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Assay Specificity

This kit is designed to detect total human IgG across all four isotypes and uses a natively purified human IgG standard. Some variability may be observed due to differences in sample isotype composition and the native standard material. For isotype-specific measurement, please use the kits listed below:
- Human IgG1: ab317543
- Human IgG2: ab202402
- Human IgG3: ab318929
- Human IgG4: ab320788

Human IgM, human IgA and human IgE were prepared at 10 ng/mL and 250 ng/mL in Sample Diluent NS and assayed for cross reactivity. No cross-reactivity was observed for IgM or IgE at either concentration with a mean OD deviation from background of -0.01. No cross-reactivity was observed for IgA at 10 ng/mL and only 0.4% cross-reactivity at 250 ng/mL of IgA. Purified human IgG was assayed at 5 ng/mL in the presence and absence of 250 ng/mL of human IgM, human IgA and human IgE to determine interference. After background subtraction, recovery of human IgG was observed at a mean of 98% with a standard deviation of 0.06.

Immunoglobulin G (IgG) serves as an essential component of the immune system functioning mainly as an antibody. It is found in blood and extracellular fluid making it one of the most prevalent antibodies in the human body. IgG weighs approximately 150 kDa and it is expressed by B cells as part of the humoral response to pathogens. Alternate names include gamma globulin and Ig gamma. IgG antibodies form a critical aspect of adaptive immunity by targeting specific antigens for neutralization or destruction.
Biological function summary

IgG participates in several immune processes by binding to antigens and forming immune complexes. It facilitates pathogen opsonization initiating phagocytosis by immune cells like macrophages and neutrophils. IgG also activates the classical complement pathway which enhances pathogen clearance. IgG exists in a monomeric form and can cross the placenta providing passive immunity to the fetus. In laboratory settings its high specificity makes it valuable in techniques like IgG ELISA for detecting antigen presence.

IgG plays an integral role in the complement system and the adaptive immune response. It partners with proteins such as C1q to initiate the classical complement pathway amplifying the body's ability to detect and remove pathogens. Constant region of immunoglobulin (Ig) heavy chains. Igs are membrane-bound or secreted glycoproteins produced by B lymphocytes. In the recognition phase of humoral immunity, the membrane-bound Igs serve as receptors, which upon binding to a specific antigen trigger the clonal expansion and differentiation of B lymphocytes into Ig-secreting plasma cells. Secreted Igs known as antibodies mediate the effector phase of humoral immunity by blocking the interaction of infectious antigens with cellular receptors (via the antigen-binding region) and eliciting effector mechanisms that lead to pathogen neutralization (via the constant region) (PubMed : 17576170, PubMed : 20176268, PubMed : 22158414). The antigen-binding region is formed by the variable domain of one heavy chain paired with the variable domain of its associated light chain. Each Ig molecule has two antigen-binding sites with remarkable affinity for a particular antigen due to V-(D)-J rearrangement, somatic hypermutations and affinity maturation of the variable domains upon antigen exposure (PubMed : 17576170, PubMed : 20176268, PubMed : 22158414). The constant region defines the Ig isotype that perform distinct sets of effector functions. B cells diversify and rearrange their Ig constant regions through class-switch recombination, a process by which the constant region is switched from one Ig isotype to another, namely from IgM and IgD to IgG, IgA and IgE (PubMed : 17576170, PubMed : 20176268, PubMed : 22158414). The constant region of Ig gamma-1 (IgG1) isotype interacts (via the fragment crystallizable, Fc) with receptors on innate immune cells and the complement system to mediate humoral effector functions, including antibody-dependent cellular cytotoxicity or phagocytosis, complement-dependent cytotoxicity and inflammatory responses.
