Interpolated concentrations of native IGFBP2 in human PBMC cell culture supernatant samples treated for 46 hours with 1.5% PHA-M.

The concentrations of IGFBP2 were measured in duplicates interpolated from the IGFBP2 standard curves and corrected for sample dilution. Undiluted samples are as follows : PBMC supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean IGFBP2 concentration was determined to be 3034.42 pg/mL in PBMC supernatant.

Interpolated concentrations of native IGFBP2 in human serum and plasma samples.

The concentrations of IGFBP2 were measured in duplicates interpolated from the IGFBP2 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 5% plasma (citrate) 5% plasma (heparin) 10% and plasma (EDTA) 5%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean IGFBP2 concentration was determined to be 257.87 ng/mL in serum 273.81 ng/mL in plasma (citrate) 139.86 ng/ml in plasma (heparin) and 118.43 ng/ml in plasma (EDTA).

How the assay works

Human IGFBP2 ELISA Kit, Chemiluminescent (ab327300) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA^® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Human IGFBP2 (ab327300) protocol summary

Multifunctional protein that plays a critical role in regulating the availability of IGFs such as IGF1 and IGF2 to their receptors and thereby regulates IGF-mediated cellular processes including proliferation, differentiation, and apoptosis in a cell-type specific manner (PubMed : 18563800, PubMed : 38796567). Functions coordinately with receptor protein tyrosine phosphatase beta/PTPRB and the IGF1 receptor to regulate IGF1-mediated signaling by stimulating the phosphorylation of PTEN leading to its inactivation and AKT1 activation (PubMed : 22869525). Plays a positive role in cell migration via interaction with integrin alpha5/ITGA5 through an RGD motif (PubMed : 16569642). Additionally, interaction with ITGA5/ITGB1 enhances the adhesion of endothelial progenitor cells to endothelial cells (PubMed : 26076738). Upon mitochondrial damage, facilitates apoptosis with ITGA5 of podocytes, and then activates the phosphorylation of focal adhesion kinase (FAK)-mediated mitochondrial injury (PubMed : 38796567).
