Interpolated concentrations of native HSPG2 in human serum, plasma samples.

The concentrations of HSPG2 were measured in duplicates, interpolated from the HSPG2 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 15%, plasma (citrate) 15%, plasma (EDTA) % and plasma (Heparin) 15%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean HSPG2 concentration was determined to be 39.48 ng/mL in serum, 27.14 ng/mL in plasma (citrate) and 76.72 ng/mL in plasma (EDTA) and 14.99 ng/ml in plasma (heparin)

Human HSPG ELISA kit (ab274393) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human HSPG protein in human serum and plasma. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human HSPG with 47.215 pg/mL sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

-Single-wash protocol reduces assay time to 90 minutes or less

-High sensitivity, specificity and reproducibility from superior antibodies

HSPG2 functions as an integral component of basement membranes. In the glomerular basement membrane, it provides a barrier that is both size- and charge-selective. HSPG2 serves as an attachment substrate for cells and plays essential roles in vascularization. It is critical for normal heart development and for regulating the vascular response to injury. HSPG2 in an anti-angiogenic and anti-tumor peptide that inhibits blood vessel growth in the chorioallantoic membrane. It blocks endothelial cell adhesion to fibronectin and type I collagen. Interaction with its ligand, integrin alpha2/beta1, is required for the anti-angiogenic properties.

Heparan Sulfate Proteoglycan 2 also known as Perlecan is a large multidomain proteoglycan with a mass of over 500 kDa. It contains heparan sulfate chains that contribute to its role in the extracellular matrix. Perlecan is expressed in various tissues including cartilage basement membranes and vascular tissues. It interacts with other matrix proteins providing structural integrity and influencing cell behavior.

Component of the glomerular basement membrane (GBM), responsible for the fixed negative electrostatic membrane charge, and which provides a barrier which is both size- and charge-selective. It serves as an attachment substrate for cells. Plays essential roles in vascularization. Critical for normal heart development and for regulating the vascular response to injury. Also required for avascular cartilage development (PubMed : 12435733, PubMed : 15591058, PubMed : 19789387). In muscle, it is essential for localizing acetylcholinesterase (AChE) at the neuromuscular junctions (NMJ), most probably acting as an adapter that links the acetylcholinesterase collagenic tail peptide (COLQ) to alpha-dystroglycan, and is therefore involved in the down-regulation of colinergic synaptic transmission (By similarity).. Endorepellin. Anti-angiogenic and anti-tumor peptide that inhibits endothelial cell migration, collagen-induced endothelial tube morphogenesis and blood vessel growth in the chorioallantoic membrane. Blocks endothelial cell adhesion to fibronectin and type I collagen. Anti-tumor agent in neovascularization. Interaction with its ligand, integrin alpha2/beta1, is required for the anti-angiogenic properties. Evokes a reduction in phosphorylation of receptor tyrosine kinases via alpha2/beta1 integrin-mediated activation of the tyrosine phosphatase, PTPN6.. LG3 peptide.
