    Abcam’s Ferritin Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Ferritin concentrations in plasma, serum, milk, cerebrospinal fluid samples


    A Ferritin specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently a Ferritin specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Complex is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of Ferritin captured in plate.


    The entire kit may be stored at -20°C for long term storage before reconstitution - Avoid repeated freeze-thaw cycles.

The proper value in human plasma is 8-400 ng/ml. On average we get 60 ng/ml for our samples.

I'd like to know some information about sample preparation for your Ferritin Human ELISA Kits.

I have to measure ferritin levels in human serum but I have to pre-treat my sera samples with a buffer.

I have to completely set up the experiments so I have not decided yet in which buffer re-suspend my samples (i.e. phosphate, borate or acetate buffer…).

Can I use your kits to measure sera sample previously re-suspended in this kind of buffers or they interfere with your ELISA assay? Are there other buffers that interfere (or not) with your ELISA assay?

Do I have to re-suspend also standard samples (or dilute my sera sample) in the same buffer I used for pre-treatment?

ab108698 Ferritin Human ELISA Kit: for pre-treatment of serum samples we recommend HEPES buffer.

ab108837 Ferritin Human ELISA KIt: I am sorry this is not tested with other buffers, so we would not be able to advise which buffer to use. Therefore, we recommend to use the buffers that come with the kit in this case.

Do you need antibodies that are specific for ferritin heavy or ferritin light chains? Looking through our catalog, I only found one that has been tested in ELISA and that was raised against total ferritin, ab7332-

Since this is a polyclonal raised against isolated nativeprotein, youcould potentially use it as the capture and detection antibody (if detection ab were conjugated). You could also use ab7332 as a detection antibody with a monoclonal capture antibody specific for heavy or light chain ferritin.

The only tested pair that we have for a ferritin protein is specific for mitochondrial ferritin, which I don't think is applicable to your studies but please let me know if this assumption is incorrect.
