Abcam's anti-Clostridium tetani toxin IgG Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of IgG class antibodies against Clostridium tetani toxin in Human serum and plasma.

A 96-well plate has been precoated with Clostridium tetani toxin antigens to bind cognate antibodies. Controls or test samples are added to the wells and incubated. Following washing, a horseradish peroxidase (HRP) labelled anti-Human IgG conjugate is added to the wells, which binds to the immobilized Clostridium tetani toxin-specific antibodies. TMB is then catalyzed by the HRP to produce a blue color product that changes to yellow after adding an acidic stop solution. The density of yellow coloration is directly proportional to the amount of Clostridium tetani toxin IgG sample captured in plate.

Clostridium tetani toxin IgG also known as tetanus toxoid antibody targets the tetanospasmin produced by the bacterium Clostridium tetani. Tetanus toxin is a notable neurotoxic protein with a molecular mass of approximately 150 kDa. The toxin is expressed primarily by Clostridium tetani bacteria in contaminated wounds or environments such as soil or animal excretion. This protein targets neurons by inhibiting neurotransmitter release which can result in motor function impairment.
Biological function summary

The role of Clostridium tetani toxin involves its activity as a potent neurotoxin causing violent muscular contractions. The toxin enters motor neurons and travels retroaxonally to the central nervous system preventing the release of inhibitory neurotransmitters like glycine and gamma-aminobutyric acid (GABA). It does not form part of a complex but independently exerts its significant effects on the nervous system.

Pathways

Tetanus toxin adversely affects normal neurotransmission processes. It specifically interrupts synaptic vesicle release pathways by cleaving synaptobrevin a critical protein for neurotransmitter vesicle fusion. This action interferes with the Wnt signaling pathway and affects proteins related to synaptic vesicle recycling including synaptotagmin and SNAP-25 further exacerbating the impairment of neurotransmission.

Clostridium tetani toxin is directly involved in tetanus a life-threatening condition characterized by muscle stiffness and spasms. The disease can lead to severe complications such as respiratory failure if untreated. Additionally the toxin's disruption of inhibitory neurotransmitters links it to motor disorders wherein imbalance in neural signaling plays critical roles. Tetanospasmin has a significant connection with neural proteins like synaptobrevin influencing symptoms associated with nerve and muscle disorders.
