Serum from eight individual healthy Chromogranin A male donors was measured in duplicate.

Interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Chromogranin A (CHGA) concentration was determined to be 58.8 ng/mL with a range of 10.4 – 171.9 ng/mL.

Interpolated concentrations of native Chromogranin A in human serum plasma (EDTA) plasma (citrate) and plasma (heparin).

The concentrations of Chromogranin A (CHGA) were measured in duplicates interpolated from the target standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 5% plasma (EDTA) 5% plasma(citrate) 5% plasma (heparin) 5%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean target concentration was determined to be 56.7 ng/mL in serum 48.5 ng/mL in plasma (EDTA) 53.0 ng/mL in and plasma (citrate) and 40.1 ng/mL in plasma (heparin).

How the assay works

Human Chromogranin A ELISA Kit, Chemiluminescent (ab326385) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Human Chromogranin A (ab326385) protocol summary

1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature
2. Wash
3. Add ChemiHRP development solution - incubate for 2 min

Pancreastatin. Strongly inhibits glucose induced insulin release from the pancreas.. Catestatin. Inhibits catecholamine release from chromaffin cells and noradrenergic neurons by acting as a non-competitive nicotinic cholinergic antagonist (PubMed : 15326220). Displays antibacterial activity against Gram-positive bacteria S.aureus and M.luteus, and Gram-negative bacteria E.coli and P.aeruginosa (PubMed : 15723172, PubMed : 24723458). Can induce mast cell migration, degranulation and production of cytokines and chemokines (PubMed : 21214543). Acts as a potent scavenger of free radicals in vitro (PubMed : 24723458). May play a role in the regulation of cardiac function and blood pressure (PubMed : 18541522).. Serpinin. Regulates granule biogenesis in endocrine cells by up-regulating the transcription of protease nexin 1 (SERPINE2) via a cAMP-PKA-SP1 pathway. This leads to inhibition of granule protein degradation in the Golgi complex which in turn promotes granule formation.
