Human CD16 ELISA Kit (ab277469) is an in-vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human CD16 in serum (human CD16 concentration is low in normal serum/plasma, and may not be detectable in this assay), plasma (Collect plasma using Citrate and Heparin as an anticoagulant. EDTA is not recommended as an anticoagulant for use in this assay), and cell culture supernatants.

This assay employs an antibody specific for Human CD16 coated on a 96-well plate. Standards and samples are pipetted into the wells and Human CD16 present in a sample is bound to the wells by the immobilized antibody. The wells are washed, and biotinylated anti-Human CD16 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Human CD16 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

CD16 also known as FCGR3A or CD16A protein is a low-affinity receptor for the Fc region of IgG. It weighs approximately 50-70 kDa and exists on the surface of several immune cells such as natural killer (NK) cells some populations of T cells and macrophages. The expression of CD16 mediates various cellular responses including antibody-dependent cellular cytotoxicity (ADCC) where it acts by engaging with immune complexes. Researchers often use anti-CD16 antibodies in flow cytometry to analyze the presence and quantity of CD16-expressing cells.
Biological function summary

This receptor plays a significant role in immune system regulation by contributing to the activation of immune effector cells. CD16 is not part of a larger complex but it operates in concert with CD3 proteins to trigger intracellular signaling pathways upon engagement with its ligand.
