    Abcam’s CD14 Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of CD14 in Human serum, plasma, buffered solutions or cell culture media.


    A monoclonal antibody specific for CD14 has been coated onto the wells of the microtiter strips provided. Samples, including standards of known CD14 concentrations and unknowns are pipetted into these wells. During the first incubation, the standards or samples and a biotinylated monoclonal antibody specific for CD14 are simultaneously incubated. After washing, the enzyme Streptavidin-HRP, that binds the biotinylated antibody is added, incubated and washed. A TMB substrate solution is added which acts on the bound enzyme to induce a colored reaction product. The intensity of this colored product is directly proportional to the concentration of CD14 present in the samples.


    This kit will recognize both endogenous and recombinant Human CD14.

Does this kit recognize soluble CD14 as well as membrane bound? According to the uniprot link below, amino acids 346-375 are removed from the mature, soluble form.

ab45523 should recognize soluble CD14, as it has been tested and is guaranteed to detect the protein in cell culture supernatant, in which only soluble CD14 would be present.

I am sorry for the delay in getting back to you.

As discussed in my previous email, the human CD14 ELISA kits ab99989 and ab45523 are both sandwich ELISA kits which bind CD14 to the surface of the plate and use another antibody to detect the bound protein. We obtain these kits from different sources so they use different antibodies and have been developed independently. They should however work in very similar ways, both kits can be used to measure CD14 in a number of different samples andboth are compatible with using with plasma as you are hoping to do. Both kits also provide a CD14 standard to quantify the levels measured.

I would also like to bring it to your attention that unfortunately we do not have the kit ab45523 in stock and this will take 1-2 weeks for delivery. The kit ab99989 would bedelivered straightaway. Thank you for your enquiry. I can suggest the following procedure which can be used to lyse the THP cells ready for CD14 detection using this ELISA kit: Lysis Buffer : Tris 50mM Nacl 150mM 1%NP40 (Nonidet P40) pH=8.0 1-Collect the cells by centrifugation at 400g for 5min. Remove the supernatant and wash once again with PBS 1X. 2- Place the tube containing the cells on ice. Resuspend cells in 1ml of lysis buffer for 10[sup]7 [/sup] cells. 3- Leave on ice for 30min with occasional mixing. 4- Carefully remove the supernatant and aliquot in several vials. (Keep at -70°C) It should be suitable for use with human THP-1 cell lysate. I hope this information is helpful. Should you have any further questions, please do not hesitate to contact us again.
