Frequently asked questions
How do chemiluminescent ELISA kits differ from traditional colorimetric ELISA kits?
Chemiluminescent ELISA kits use a luminescent substrate that emits light as a signal, which is then measured via a luminometer. This method generally provides higher sensitivity and a broader dynamic range compared to the colorimetric method, which relies on a chromogenic substrate and optical density measurements.
Are these kits suitable for all types of samples?
Chemiluminescent ELISA kits can be used with a variety of sample types, including serum, plasma, cell culture supernatants, and other biological fluids. However, it's important to consider a specific kit's compatibility with your sample type for optimal results.
What equipment is needed to use a chemiluminescent ELISA kit?
You will need a plate reader equipped to detect chemiluminescence.

Evaluating a new assay shouldn't require a large upfront commitment.
If you're new to SimpleStep Ignite™ ELISA, you can get 50% off your first kit, making it easier to assess performance in your own samples and workflows before wider adoption.
Validate with confidence
Switching assays mid-project can feel like a risk. That's why SimpleStep Ignite™ ELISA was designed to make validation, adoption, and scaling as straightforward as possible. The combination of enhanced sensitivity, broad measuring range, and a familiar one-wash workflow helps you transition with confidence while minimizing disruption to ongoing work.
What is a chemiluminescent ELISA?
Chemiluminescent ELISA kits use light-emitting reactions to offer highly sensitive and rapid detection of biomarkers.
These kits use a chemiluminescent substrate to produce light in response to antigen-antibody interactions, which allows for greater sensitivity and broader dynamic range than traditional ELISA. Our 90‑minute colorimetric SimpleStep® ELISA kits have already saved researchers around the world 82 years at the bench. SimpleStep Ignite™ ELISA builds on this legacy - helping accelerate the pace of scientific discovery.
Faster results
Improved assay performance should not require longer workflows.
While many commercially available ELISA assays require several hours to complete, SimpleStep Ignite™ ELISA delivers results in just 55 minutes using a single-incubation, single-wash protocol.
- Faster experimental turnaround
- Reduced hands-on time
- More assays completed per day
- Accelerated decision-making
- Improved laboratory productivity

Broader measuring range
More measurable results. Fewer reruns.
A broader measuring range allows accurate quantification across a wider concentration spectrum, reducing the need for repeated testing and sample dilution.
We compared reported Human C-peptide concentrations from 450 samples and found that all fell within the measuring range of SimpleStep Ignite™, demonstrating its ability to support first-run success across a broad range of sample concentrations.
This helps increase the likelihood that more samples can be measured first time, supporting a smoother workflow with fewer repeats and less hands-on time.
- Wider dynamic range reduces out-of-range results
- Fewer re-tests lead to faster data delivery
- Improved success rate for unknown sample concentrations

Higher sensitivity
Detect low-abundance analytes with confidence.
Across multiple assays, chemiluminescent readout improves the signal-to-noise ratio, increasing analytical sensitivity and precision (Figure 1).
The data in the graphs show enhanced signal-to-noise performance and stronger detection at lower analyte concentrations, helping researchers achieve reliable quantification of low-abundance targets.
Chemiluminescent detection can achieve substantially greater sensitivity than conventional colourimetric approaches, making it particularly valuable when sample availability is limited or target concentrations are low (Figures 2a and 2b).

Comparison of the signal-to-noise ratios for Human IL‑4, measured by chemiluminescent (orange) and colorimetric (blue) detection. Chemiluminescent readout improves low‑end signal-to-noise, thereby increasing analytical sensitivity and precision
Traditional ELISA methods often require compromises between sensitivity, assay time and measuring range.
SimpleStep Ignite™ ELISA kits are designed to help overcome these challenges. The examples below demonstrate how chemiluminescent detection improves assay performance, delivering enhanced signal-to-noise ratios and greater sensitivity across multiple targets compared with conventional colourimetric ELISA.
