    Wherever possible, you should prepare and use solutions on the same day. However, if you need to make up stock solutions in advance, we recommend that you store the solution as aliquots in tightly sealed vials at -20°C.     ab32034 staining p27 KIP1 in MCF7 cells treated with NS 398 (ab120295), by ICC/IF. Increase in p27 KIP1 expression correlates with increased concentration of NS 398, as described in literature.
    The cells were incubated at 37°C for 6h in media containing different concentrations of ab120295 (NS 398) in DMSO, fixed with 4% formaldehyde for 10 minutes at room temperature and blocked with PBS containing 10% goat serum, 0.3 M glycine, 1% BSA and 0.1% tween for 2h at room temperature. Staining of the treated cells with ab32034 (1/100 dilution) was performed overnight at 4°C in PBS containing 1% BSA and 0.1% tween. A DyLight 488 goat anti-rabbit polyclonal antibody (ab96899) at 1/250 dilution was used as the secondary antibody.

Thanks. I am also currently looking for a VEGF inhibitor and mostly they are for the receptor. Any chemical inhibitor in line with NS-398 should work fine for us. May be if I get one, I will let you know.

That is the problem with this target. I have been able to find chemical inhibitors of VEGFR but not for VEGF. There seems to be mainly antibody inhibitors of VEGF. If you do find a compound you are interested in, please do let me know and I can see if we could consider adding it to the catalogue.

I am looking for primary HO-1 antibody (Haem oxygnase) and COX-2 (mono or polyclonal , raised in rabbit against mouse, rat and human, applicability). Can you suggest some good ones. Also, I am looking for chemical inhibitors for COX-2 (NS-398) and VEGF (I actually don’t know the name of the inhibitor), can you tell me the availability of the same?

I hope your grant application is going well. We have several antibodies against the targets you have mentioned which may be of interest to you:

HO-1

I would suggest looking at the rabbit polyclonal https://www.abcam.com/Heme-Oxygenase-1-antibody-ab13243.html. This has been shown to react with human, mouse and rat samples and has been used in ICC, IHC-Paraffin embedded, sELISA and Western blotting to date. We also have a very popular mouse monoclonal https://www.abcam.com/Heme-Oxygenase-1-antibody-HO-1-1-ab13248.htmlwhich has also been used with human, mouse and rat samples as well as in ELISA, Flow Cytometry, ICC,IHC-Frozen sections, and Western blotting. Bit I think you were more interested in antibodies raised in rabbit.

COX-2

For this target I would suggest having a look at the rabbit polyclonal https://www.abcam.com/COX2--Cyclooxygenase-2-antibody-ab15191.html. This has again been tested with mouse, rat and human samples and has been used in ICC/IF, IHC-Frozen sections, IHC-Parrafin embedded, IP, sELISA and Westernblotting.

We also have the NS-398 compound (COX-2 inhibitor) which is available under the catalogue number https://www.abcam.com/NS-398-ab120295.html.

I am afraid I have not been able to find a similar inhibitor of VEGF. Do you have a particular compound in mind? It may be that I can suggest we start stocking it.
