    Wherever possible, you should prepare and use solutions on the same day. However, if you need to make up stock solutions in advance, we recommend that you store the solution as aliquots in tightly sealed vials at -20°C. Generally, these will be useable for up to one month. Before use, and prior to opening the vial we recommend that you allow your product to equilibrate to room temperature for at least 1 hour.

    ab55051 staining GABA B receptor 1 in SK-N-SH cells treated with NMDA (ab120052), by ICC/IF. Internalization of GABA B receptor 1 correlates with increased concentration of NMDA, as described in literature.
    The cells were incubated at 37°C for 30 minutes in media containing different concentrations of ab120052 (NMDA) in DMSO, fixed with 4% formaldehyde for 10 minutes at room temperature and blocked with PBS containing 10% goat serum, 0.3 M glycine, 1% BSA and 0.1% tween for 2h at room temperature. Staining of the treated cells with ab55051 (1 µg/ml) was performed overnight at 4°C in PBS containing 1% BSA and 0.1% tween. A DyLight 488 goat anti-mouse polyclonal antibody (ab96879) at 1/250 dilution was used as the secondary antibody. Nuclei were counterstained with DAPI and are shown in blue.

Question

To whom it may concern, We use NMDA to induce a cognitive deficit in an passive avoidance task in rat. I have used NMDA from sigma-aldrich (product code XXXX) before which is in a free aminoacid form. Now we are trying your NMDA which is in a hemihydrate form (ab120052) and we have some problems. First it is hard to disollute (15 min in ultrasonic bath, 42 mg/1ml water) in comparsion to the sigmas NMDA. Second, the rats behave different. Abcam NMDA seems to be stronger and some rats have epileptoformic seizures after systemic administration (42 mg/kg i.p.). We have observed also some mortality, approx. 10 %. With Sigma NMDA we did not observe any seizures or mortality. Do you have any idea why it is happening ? May I kindly ask you for NMR spectra of this batch ? with best regards

Thank you for contacting us.
I am sorry to hear you are not observing the expected results with the reagent supplied.
It is possible that ab120052 acts more potently due to it having a higher purity when compared to Sigma’s. I have attached the NMP spectra for ab120052. If you had Sigma’s NMR spectra for this product, we would be happy to compare it against ours to determine if this is the case.
It is also worth considering that variations in the strength of the epileptoformic seizures can occur due to variations in batches of rats.
We would also recommend using the batch specific molecular weight when calculating doses.
As a final note, variations in seizure strength could occur if the solvent/saline solution is not fresh or contaminated – this should be checked before carrying out experimental procedures.
