Alpha 1 Adrenergic Receptor commonly referred to as ADRA1 is a G-protein coupled receptor with an approximate mass of 60 kDa. It mainly resides in smooth muscle cells throughout the vascular system and also in tissues such as the heart and skin. ADRA1 activates through the binding of catecholamines like norepinephrine leading to subsequent cellular responses. This receptor exhibits at least three different subtypes: ADRA1A ADRA1B and ADRA1D each with distinct roles and expression patterns.
Biological function summary

Alpha 1 Adrenergic Receptor contributes to a wide array of physiological activities including smooth muscle contraction vasoconstriction and pupil dilation. The receptor does not function in isolation but integrates into signaling complexes that modulate intracellular second messenger systems. As a receptor engaged in various critical physiological functions its expression and activity need precise regulation to ensure homeostasis and effective organismal responses.

Pathways

Alpha 1 Adrenergic Receptor is deeply entwined in the adrenergic signaling pathway which significantly impacts the cardiovascular and nervous systems. This receptor activates the phospholipase C pathway resulting in the production of inositol triphosphate (IP3) and diacylglycerol (DAG) which ultimately open calcium channels in the endoplasmic reticulum. Through these pathways ADRA1 interacts with other proteins such as ADRA2 and beta-adrenergic receptors contributing to a balanced sympathetic nervous response.

Alpha 1 Adrenergic Receptor plays a role in conditions like hypertension and benign prostatic hyperplasia (BPH). The receptor's involvement in these diseases often relates to its function in vascular and smooth muscle regulation. Overactivation of ADRA1 may contribute to vasoconstriction leading to hypertension while in BPH it can promote muscle contraction around the prostate. Related proteins such as ADRA2 and phospholipase C have also shown implications in these disorders through similar or complementary pathways.
