      Inhibits 5-HT binding to VMAT2 (IC50 = 1 µM (comparable to dopamine itself)). Released via exocytosis in the striatum in mouse acute slice preparation. Labels dopamine and other presynaptic terminals in the striatum.
      Compatible with GFP tags and other optical probes. Sufficiently bright, photostable and suitable for two-photon fluorescence microscopy.

      Store at +4°C. Store under desiccating conditions. The product can be stored for up to 12 months.

      Soluble in DMSO to 100 mM and in ethanol to 100 mM
    • 使用に関する注意

      Wherever possible, you should prepare and use solutions on the same day. However, if you need to make up stock solutions in advance, we recommend that you store the solution as aliquots in tightly sealed vials at -20°C. Generally, these will be useable for up to one month. Before use, and prior to opening the vial we recommend that you allow your product to equilibrate to room temperature for at least 1 hour.

      To make a 10 μM solution for slice incubation, 25 μl of a 20 mM FFN511 stock solution in DMSO is added to 50 ml of ACSF. This results in a 10 μM FFN511 solution in ACSF containing 0.05% DMSO.

    Thank you for contacting us and your interest in our products.

    The information we have in relation to the emission and excitation of FFN511 is from the following reference:

    Gubernator NG et al. Fluorescent false neurotransmitters visualize dopamine release from individual presynaptic terminals. Science 324:1441-4 (2009). (PubMed: 19423778)

    Although the emission and excitation maxima for FFN511 are 406 nm and 501 nm respectively (in pH7 buffer), the fluorescence measurements taken by Gubernator et al. was using two-photon excitation microscopy. The excitation was performed using a multiphoton laser scanning microscope with a Titanium-sapphire laser with excitation of 760 nm applied and the emission measured using 480-520 nm band pass filter.

    Thank you for your inquiry.

    We do ot have any information about the pKa value of FFN511. The available literature also does not seem to mention the pKa value of FFN511.

    In relation to the pH range, the only mention is in the supplemental information

    Gubernator NG et al. Fluorescent false neurotransmitters visualize dopamine release from individual presynaptic terminals. Science 324:1441-4 (2009).(PubMed: 19423778)



    ,is that it was used at pH 7.4. I would assume this is due to it being a physiologically relevant pH.

    As we do not currently undertake our own research I can offer you advice based on information available in a recent paper by Gubernator et al., 2009. I can confirm that excitation of FFN551 occurs at 760 nm, with optimal fluorescence of striatal terminals seen through a band pass filter (emission 480-520 nm). I have attached a copy of the paper for your information. I have also attached a copy of the supporting material where there is more detailed information regarding the
    photo physical properties of FFN511.
