    Protein Carbonyl Content Assay Kit ab126287 is designed to provide a simple and accurate method of quantifying carbonyls in protein samples.


    The protein carbonl assay protocol is based on the reaction of DNPH with protein carbonyls. This is the most popular assay type for measuring protein carbonyl content. DNP hydrazones formed in this reaction are easily quantifiable at 375 nm absorbance.


    When used with, for example a 1 mg (~15 nmol) sample, this protein carbonyl assay protocol has a detection limit of about 0.15 nmol carbonyl. For context, BSA typically contains approximately 1-3 nmol carbonyl/mg.


    Protein carbonyl assay protocol summary:
    - add DNPH to samples and incubate for 10 min
    - add TCA to samples, incubate for 5 min, spin for 2 min, and discard supernatant
    - wash pellet by sonicating in acetone, spin and remove acetone, repeat acetone wash step
    - add Guanidine solution and resolubilize pellet, and transfer to plate
    - analyze with a microplate reader to measure protein carbonyl content, also perform a protein assay (eg. BCA assay) on samples to establish protein concentration

    This product is manufactured by BioVision, an Abcam company and was previously called K830 Protein Carbonyl Content Assay Kit. K830-100 is the same size as the 100 test size of ab126287.

From what I understand from the description, the Oxyblot seems to detect carbonyl groups introduced into proteins by oxidative reactions.
We have the assay kit:
https://www.abcam.com/protein-carbonyl-content-assay-kit-ab126287.html
It detects carbonyl groups in protein samples.
So it looks to me like it's quite similar, but maybe you have to tell the customer to have a look at the protocol and see if that's what he wants.
If the customer would like to test this kit on untested samples, I could offer them Abcam's Abtrial.

Hello, Iam very intersting in the dealing with your product in the abcam company, but I have a question about your product ( ab126287) Protein carbonyl content Assay Kit, the wave length that I must use it to measure optical density is ˜3750 and this according to the instruction in this kit, my problem is the readers in my university dont have the filter for this wavelength, the most closest one 405 wave length so my question is can I use 405 wave length to read the result for this Kit (ab126287).

please I need the answer as soon as possible

Every plate reader has flexibility in the band width detection range. If the reader being used has a flexibility of ± 30, you can use 345 to 405 nm for the 375 nm recommendation. Otherwise it might compromise the efficiency of the detection.
