    GST Assay Kit (Colorimetric) (ab65326) is used to detect GST activity in crude cell lysates and purified protein fractions, and also to quantitate GST-tagged fusion proteins.


    The GST assay protocol is based on the GST-catalyzed reaction between GSH and the GST substrate CDNB.


    CDNB has the broadest range of isozyme detectability (e.g., alpha-, mu-, pi-, and other GST isoforms). The GST-catalyzed formation of GS-DNB produces a dinitrophenyl thioether which can be detected by spectrophotometer at 340 nm.


    One unit of GST activity is defined as the amount of enzyme producing 1 µmol of GS-DNB conjugate/min under the conditions of the assay. Detection limit of assay: active GST < 1 mU.


    GST assay protocol summary:
    - add samples and positive control to wells
    - add glutathione to wells
    - add reaction mix
    - analyze with a microplate reader for at least 10 min

    This product is manufactured by BioVision, an Abcam company and was previously called K263 GST Colorimetric Activity Assay Kit. K263-100 is the same size as the 100 test size of ab65326.

    Glutathione S-transferase (GST, EC 2.5.1.13) is a family of enzymes that play an important role in detoxification of xenobiotics. GST catalyzes the formation of the thiol group of glutathione to electrophilic xenobiotics. It utilizes glutathione to scavenge potentially toxic compounds including those produced as a result of oxidative stress and is part of the defense mechanism against the mutagenic, carcinogenic and toxic effects of such compounds.

Meine Frage bezieht sich auf Ihre Produkte
Alanine Transaminase Activity Assay Kit (ab105134)
und GST Activity Assay Kit (Colorimetric) (ab65326).
Ich möchte diese Kits nutzen, um die beiden Enzyme im Überstand meiner
Zellkulturen nachzuweisen.
Auf Ihrer website wird für diese beiden Produkte angegeben, dass sie
für den "Probentyp" Zellkulutrmedium bzw. Zellkulturüberstand geeignet
sind. Leider finde ich in der jeweiligen Anleitung (protocol booklet)
keine Angaben zur Behandlung von Zellkulturüberstand.
Darum meine Frage: Ist es sicher, dass ich diese Kits zum Nachweise
einer Enzymfreistzung ins Zellkulturmedium verwenden kann?

Ich möchte hiermit auch bestätigen, dass beide Kits für Zellkulturüberstand getestet und auch garantiert sind.

Sie haben Recht, dass eine Probenaufbereitung nicht gesondert erwähnt wird.

Zellkulturüberstand (frei von Zellen) kann direkt eingesetzt werden. Wir empfehlen einen unverdünnte Probe und dann einige verschiedene Verdünnungen zu testen, damit man sicherstellt, dass die Werte in den Bereich der Standardkurve fallen.

Thank you for your enquiry and your interest in our products.

This kit is not specific for any GST isoforms so it detects the total GST enzymatic activity in the samples (using CDNB substrate).

There are some other GST substrates available but most of these compounds have overlapping specificity to the different forms. Your customer may be able to find some published articles on this topics using PubMed::

I thought I had it figured out by heating the GSH for solubility, but then, after running a plate or two successfully, the next plate was really bad. Anyway, I may take you up on your offer for extra GSH and/or buffer, but for now, we are thinking of just looking at other endpoints instead. I will be in touch when I know more.

That is fine. In the next kit I used, the powder seemed drier upon thawing, and it dissolved almost immediately into the buffer without much mixing (and no heating) at all. The other vials of lyophilized GSH (the ones that did not dissolve well) looked as if they had been exposed to moisture, because I could see some liquid droplets inside the vial. Perhaps these vials were just not well sealed or something? I don't know if this could have affected solubility or not, but maybe this was the reason for some of the variability in my assays.

I have not yet check the vials that came with the remaining kits to see if any of them also appear to be affected, but I will later today and will let you know. I'm not sure what could be done about it. Maybe you will hear back from support soon with an answer.

Hi,

Yes, the positive control is also quite variable. For the samples I ran yesterday (after I emailed you), I heated the GSH mixture to 37degC for about 15 min, and then vortexed a bit more. This seemed to help tremendously. At first I was afraid to heat it, since the reagents seem temperature dependent according to the kit, but it seemed to work out fine. There was still some powder undissolved, though, and I was careful not to pipet up the insoluble debris. Luckily that plate was not full, so I did not need the full volume. However, since the first 2 kits I used this week returned unusable data, I will need to use the full volume of GSH in the rest of the kits, as I hope to squeeze in the samples that need to be run.

The units calculated for the GST activity should be in μmol/min/ml. The μmol unit comes from the extinction coefficient. I could not find any reference in out protocol to nmol. Where have you found this? I'd like to makesure there is not a mistake in the protocol.

I promise I am almost finished with questions ;)

Do you know how reliable small changes in Vmax are, or what levels above baseline are reliable?

For my PBMC samples, I am getting small Vmax values ranging from 1.7-5.0, with baseline subtracted. Are these values reliable since they are so close to baseline?

I'm trying to see if a certain treatment changes GST activity, and I don't know if a change from 2.3 to 3.5 is actually enough to consider a change.

Sorry for the delay in getting back to you.

I now have feedback in regards to the calculations you are trying to perform:

The source of the kit has confirmed that thesecond of your calculations accordingis the closest to being correct. Here is theircalculation:

4.9 x 0.105 divided by 0.0096 x 1000 x 0.2893 x 0.003 =0.5145 by 0.0083318 = 61.75 umol/min/ml.

Thedilution factor was taken as 1 because you areadding pure undiluted 3 ul of the protein into the wells. Its addition with the buffer and the mix is accounted for in the 0.105 ml calculation.

Now each ml ofyour samplewill have a total of 6 mg of protein (6ug/ul = 6 mg/ml).

Therefore the reading is 61.75 umol/min/6mg. Which is equivalent to 10.29 umol/min/mg.

Additionally, wethink that the2.3 to 3.5 “Vmax” changeshould be significant if you put it into calculations. I would make sure to do enough repeat readings to have confidence in the figures and that it is not simply down to sample variations.

Also, if you could provide me with your order number (or purchase order number) I can arrange to reimburse you for the value of a kit. Would you prefer for this to be in the form of a credit note (to be used against future Abcam orders) or as a refund?

I need to know, because I would like to use the same cell lysates for the GST activity assay as well as another assay, but I do not know if the assay buffer will be compatible with the other assay. Normally for the other assay, I use an IP lysis buffer with protease inhibitors. Any information you could give me on the assay buffer would be great.

Thank you for getting back to me and sorry for the delay in my reply.

Hopefully I can now sort out dome of the confusing information. There has been an error in the equation provided to calculate GST. Unfortunately, the extinction coefficient was entered incorrectly and should be 0.0096 umol-1cm-1, not 0.0023 as it was changed to in the most recent booklet. The 0.2893 was added to include the pathlength of the solution. The units are in µmol/min/ml. This information will be updated in our protocol as soon as possible. I am very sorry for the inconvenience this confusion has caused. I hope the equation now makes sense to you. Asan apologyfor the confusion we have caused youI would like to offer you a free GST Activity Assay kit in compensation. Please let me know if you would like for me to arrange this.

In answer to the change in GST assay buffer in the kits. The buffers are different, this change was made to improve the kit. However, the results from the previous version of the kit are going to be comparable with this new version. Please do not interchange the components from one version to the other and you should be fine. I try to get the information regarding the composition of the GST buffer for you but I think it will probably be proprietary information but I will let you know either way.

I have also arranged for the components you have asked for to be added to our catalogue. These are available under the following catalogue numbers:

In answer to your question of how you can convert the activity to relate to the amount of protein I can answer the following:

If you are starting with 1 mg of protein in 100 ul the concentration would be 0.1 mg/ml or 0.1 ug/ul, Then if theyou aretaking 0.006 ml which is 6 ul, and you are therefore starting with with 0.6 ug of the protein. The final volumes you dilute it to then does not matter, as long as this is the working solution. You should therefore not need to multiply any of the dilution values.
