alpha-L-Fucosidase Assay Kit (ab272525) is a non-radioactive, colorimetric assay based on the cleavage of 4-nitrophenol from the synthetic substrate. Nitrophenol becomes intensely colored after addition of the stop reagent. The increase in absorbance at 405 nm after addition of the stop reagent is directly proportional to the enzyme activity.

High sensitivity and wide linear range. Linear detection range (10 μL sample): 1 to 100 U/L for a 20 minute reaction.

Homogeneous and simple procedure. Simple add-mix-read procedure allows reliable quantitation of fucosidase activity within 20 minutes.

Robust and amenable to HTS. All reagents are compatible with highthroughput liquid handling instruments. Can be readily automated to measure thousands of samples per day.

The targets FUCA1 and FUCA2 known as alpha-fucosidase and alpha-L-fucosidase respectively are enzymes that perform the removal of fucose residues from glycoproteins and glycolipids. The FUCA1 protein has a molecular mass of approximately 55 kDa while FUCA2 is slightly larger at around 60 kDa. Both enzymes are expressed in various tissues throughout the body including the liver kidney and placenta. Their activity is essential for the proper degradation of fucose-containing molecules within lysosomes.
Biological function summary

These enzymes participate in the catabolism of fucose an important monosaccharide component of N-glycans glycosphingolipids and other fucosylated molecules. FUCA1 and FUCA2 are not known to be part of larger protein complexes but their role is important in the processing and recycling of fucosylated molecules. By maintaining the balance of fucose turnover these enzymes support various cellular functions and contribute to metabolic homeostasis.

Pathways

FUCA1 and FUCA2 are involved in the glycosphingolipid biosynthesis and degradation pathways.
