Me comentan mis compañeros que aunque este kit nunca se ha testado con tejidos, dada su versatilidad con distintos tipos de muestras, es muy posible que también funcione con homogenado de cerebro.

En caso de usar el kit en este tipo de muestra, os aconsejo diluir las muestras al menos 5 veces con Assay Diluent (Item E) para minimizar los efectos de los detergentes en el buffer de lisis. Es muy posible que requieran mayor dilución, pero tendrá que determinarse empíricamente durante el experimento.

Para el lisado original lo ideal sería comenzar por 1 mg/ml de proteína, o si es posible mas, para alcanzar, una vez diluido, 50-500ug /ml. De nuevo, este rango debe ser tomado como punto de partida y tendrá que ser optimizado empíricamente.

Para preparar el buffer de lisis, quizás os puedan ayudar los siguientes consejos:

En general un buffer de lisis debe cumplir los siguientes requisitos:

a) Bajo contenido en sal (700 mM o menos).

b) No contener azida sódica.

c) No contener >0.1% SDS.

d) No contener > 10mM de agentes reductores (BME o dithiothreitol).

Esto incluiría cualquier buffer usado par a IP, incluido RIPA buffer.
