According to the Swiss Prot reference for mouse the target is “strongly expressed in spleen”, this was the positive control that was used. The predicted molecular weight, according to the Swiss Prot entry would be 23.5 KDa. You are correct that the KDa of the light chain of IgG is approximately 20 KDa, however it is unlikely that interference would be seen from the IgG produced by B cells, it is more likely that the entire KDa of the IgG1 would be seen at approximately 146 KDa. A number of secondary Abs are available: If the secondary Ab is Fab specific it will cross react with both the heavy and the light chain of the Ab structure. If working with tissues or cells that have Fc receptors (spleen) a certain amount of cross reactivity will be seen unless a F(ab )2 is used to eliminate non-specific binding through Fc receptors present on cells. Alternatively, Fc receptors can be blocked. Perform an absorption step, using purified IgG from the host species of the your secondary antibody. In that case an F(ab )2 secondary antibody may not be necessary. Alternatively kits are available that are capable of directly biotinylating the primary monoclonal Ab that is being used (e.g. Dako) and thus eliminating the need to use a anti mouse secondary. Western blot analysis was carried out on PGRP-S in tissue lysates from mouse spleen using Ab13903 at a dilution of 2 µg/ml.
