Normal human Bronchial epithelial cells(NHBE) were infected with RSV and cultured for 48 h, then the virus-induced cell surface PAF receptor was blocked by incubating the cells at 37°C for 2 h with different concentrations (5ug to 10ug) of anti-PAFr rabbit polyclonal antibody (ab104162). Cells were washed and then incubated with Streptococcus pneumoniae serotype F19 for 2 h at 37°C in 5% CO2. After 2 h, cells were washed for 5 times with PBS and trypsinised with 0.025% Trypsin, and cells were serially diluted and cultured on 5% sheep blood agar plate (Oxoid, USA) at 37 C with 5%CO2, at the same time cells were counted for each well-using cytometer. Colonies were normalized for each sample with a number of cells.
At 5ug concentration, PAFr antibody did not block S.pneumoniae
adhesion but at. 10ug/ml, PAFr blocked the adhesion of S.pneumoniae.
