      Mediates the Na(+)-independent uptake of organic anions such as pravastatin, taurocholate, methotrexate, dehydroepiandrosterone sulfate, 17-beta-glucuronosyl estradiol, estrone sulfate, prostaglandin E2, thromboxane B2, leukotriene C3, leukotriene E4, thyroxine and triiodothyronine. May play an important role in the clearance of bile acids and organic anions from the liver.

        Immunocytochemistry/Immunofluorescence analysis of HepG2 cells labeling OATP1B1 (green) with ab15542 at 1/20. F-Actin staining with Phalloidin (red) and nuclei with DAPI (blue). Cells were fixed with formaldehyde and incubated with the primary antibody overnight at 4°C. A DyLight 488-conjugated secondary antibody was used. 60X magnification. Right - negative control.

      Thank you for your enquiry. I am sorry to hear that you have been having difficulties with this antibody. I have read through your technical questionaire and I have a few comments. OATP2 antibody [MDQ] (ab15442) has been tested for its applications of immunofluorescence, immunohistochemistry (Frozen sections) and immunoprecipitation. To our knowledge we are not aware of how this antibody behaves in western blotting. Therefore unfortunately we cannot guarantee it for this purpose. I am sorry to hear that you have observed differences between the batches of antibody that you have used. This is a monoclonal antibody and I would not expect different antibody preparations to have significant variation in their quality. However, occasional batch to batch variation does exist. Therefore given this I would like to recommend that you perform the optimisation to your protocol; Firstly I would like to suggest that you verify the integrity of your sample and preparation by performing a loading control immunolabelling using an antibody that targets a housekeeping protein such as GAPDH or alpha tubulin. Following on I would like to suggest that you significantly reduce the dilution of the antibody to ~1:20 in view of the antibody dilutions recommended for immunofluorescence and perform an overnight incubation at 4oC using BSA as a blocking agent. This serves to improve the sensitivity of the antibody.

      Thank you for your enquiry. Both antibodies were tested in IP using human liver samples (~20-50 ug) and about 1 ug/ml of antibody used. For IF, human liver and transfected HEK293 whole cell lysates were used. Ab15442 was used at a dilution of 1:20. The fixative was 4% PFA for ~10-15 minutes. Ab15441 was used at 1:100 in IF.       customer has blasted the peptide seq on the datasheet and thinks it is actualy the sequence for OATP-C and doesnt correspond to OATP8 at all, I checked swissProt and I concur, unless i'm missing something.

      Thank you for bringing this to our attention. The immunogen sequence is from OATP2, however this product will also recognise OATP8. The data sheet has now been updated to reflect this.
