    Normal goat serum ab7481 is used extensively for the blocking of non-specific antibody binding in tissue and cell staining, and in other applications of antibodies.

    The goat serum blocks the binding of Fc receptors in the sample to the primary and secondary antibodies used in the experiment, and also blocks non-specific binding of the antibodies to the sample.

    Typically the serum used for blocking is from a different species than the species in which the primary antibody was raised. Often the blocking serum is from the species in which the secondary antibody was raised. 

    Serum can be used directly for blocking, or as a constituent of a blocking buffer.

    Cells grown in 8-well chamber slides to 70% confluence were fixed with 4% paraformaldehyde (15 min at room temperature (RT)), and permeabilized with 100% ice cold methanol (10 min at 20°C). The slides were incubated with 10% normal goat serum (ab7481) in PBS for 1 hr, and incubated with anti-maspin (1:100) antibody alone or in a combination with either anti-lamin B (1:50), anti-HDAC1 (1:50) or anti-GRP78 (1:50) at 4°C overnight. Cells were washed and incubated for 2 hrs at room temperature (RT) with Alexa Fluor 488 (1:500) alone or in combination with Alexa Fluor 594 (1:500). The nuclei were counterstained with DAPI.
    DU145 cells infected with adenovirus expressing either maspinWT or maspinD346E Confocal immunofluorescence imaging of maspin (green) and nuclear envelope marker lamin B (red) in DU145 cells after infection.

    Slides were pretreated with Hydrogen Peroxide Block followed by 15% normal goat serum (Abcam, Cambridge, UK) for 1 hour. The primary antibody, anti-PTX3, N-terminal antibody produced in rabbit was diluted 1:300 in PBS with 2.5% goat serum (ab7481), applied to slides and incubated for 3 hours at 4°C. The primary antibody was omitted in the negative controls. The PTX3 binding was revealed using a universal secondary antibody polymer formulation conjugated to horseradish peroxidase (HRP). The HRP activity was subsequently visualized with diaminobenzidine (DAB) substrate/chromogen and counterstaining with hematoxylin was performed.

    There is no need to heat-inactivate serum if you are using it as a blocking reagent before staining fixed cells or tissue. Heat-inactivation is only necessary if you are using the serum for tissue culture. (The heat inactivates complement, and this eliminates complement-mediated cell lysis).

    ab138478 is sterilized, whereas ab7481 is not. That is the only difference between the two products and the reason that ab138478 is more expensive.

    ab7481 was not experimentally tested as blocking agent in IP procedures. But I am confident that it should be suitable in pre-clearing steps etc in the IP procedure. Please follow this link to a protocol for IP:

    On the datasheet of these sera I have a concentration of 86mg/ml for the goat serum (8,6% concentration) and 71mg/ml for the rabbit serum (7,1% concentration). So no chance I can use them at 10% for IHC blocking on free-floating sections?

    Answer

    Those values are for whole protein value which we supply as a customer service. The total protein value is not considered when making blocking solutions as the percentage used is of the whole serum. Therefore to create a 100 ml or a 10% IHC block you would use 10 ml of serum in 90 ml of diluent.

    According to the standard protocol we have for free floating sections, we recommend to incubate the sections with 0.1M PB/0.3% triton/1% blocking serum for at least 1 h.

    Answer

    To use this serum for IHC in the blocking step, it does not have to be heat treated. The heat treatment is done to inactivate complement in FCS meant for cell culture for example.

    I recommend to aliquot and freeze the serum for storage.

    Answer

    Serums will vary in how they are prepared, but there are no specific "grades" that we are aware of. Please note that when using serum as a blocking agent, there is a chance that the animal may have produced antibodies to the antigen in question and if present, the antibodies may bind to the antigen and prevent the primary antibody from binding. If this happens, just switch blocking agents.

    We do not measure the total IgG in our goat serums, but generally would estimate total IgG to be ~5-10% of the total serum protein. Concentration of total serum protein is 97mg/ml by Refractometry.
