Extracellular ligand for the NTRK1 and NGFR receptors, activates cellular signaling cascades through those receptor tyrosine kinase to regulate neuronal proliferation, differentiation and survival. Inhibits metalloproteinase dependent proteolysis of platelet glycoprotein VI (PubMed:20164177).

    Immunohistochemical staining of paraffin embedded human cerebral cortex with purified ab52918 at a working dilution of 1 in 250. The secondary antibody used is ab97051 Goat Anti-Rabbit IgG H&L (HRP) at a dilution of 1/500. The sample is counter-stained with hematoxylin. Antigen retrieval was perfomed using Tris-EDTA buffer, pH 9.0. PBS was used instead of the primary antibody as the negative control, and is shown in the inset.

    Immunofluorescence staining of U87-MG cells with purified ab52918 at a working dilution of 1 in 300, counter-stained with DAPI. Tubulin was stained with mouse anti-tubulin at a dilution of 1/1000 (ab7291) and Alexa Fluor® 594 goat anti-mouse at a dilution of 1/500 (ab150120) . The secondary antibody was ab150077 Alexa Fluor® 488 goat anti rabbit, used at a dilution of 1 in 500. The cells were fixed in 4% PFA and permeabilized using 0.1% Triton X 100. The negative controls are shown in the bottom middle and right hand panels - for the first negative control, purified ab52918 was used at a dilution of 1/200 followed by an Alexa Fluor® 555 goat anti-mouse antibody at a dilution of 1/500 and for the second negative control mouse primary antibody (ab7291) and anti-rabbit secondary antibody (ab15007) were used.

    Immunohistochemical staining of NGF mouse tissue sections (formalin/ PFA-fixed paraffin-embedded tissue sections) with unpurified ab52918. The sections were formaldehyde fixed, subjected to heat mediated antigen retrieval and blocked for 10 minutes at 25°C. The primary antibody was diluted 1/50 and incubated with the sample for 1 hour at 25°C. An HRP polymer anti-rabbit IgG system was used undiluted, as the secondary antibody.

Hi,
I use the NGF antibody ab52918 from Abcam for IHC. To check the specificity of the staining i would like to obtain the immunising peptide and perform blocking of the antibody with immunising peptide. I can find the peptide on the abcam website.
Could you please let me know if abcam sells this peptide and if not could they provide the name of a company that does?

Question

Hi,

I have a customer enquiring about the concentration of ab52918. They have a rabbit monoclonal IgG which they wanted to use as isotype control and need to match its conc with that of the primary antibody.

I pointed out to them that this antibody is a “tissue culture supernatant” and hence the conc of the specific antibody is not known. They mentioned that:

'when i order Rb mAbs from cell signaling Technology they are able to tell me concentration (although not written on datasheet) and do not state any purification step in their datasheet either.'

i still think abcam could possibly know concentration of IgG in the supernatant - otherwise how would they do quality control and adjust for lot to lot variations?

They also want to know:

'does Abcam have a rabbit mAb culture supernatant type that they advise to use as negative control then?'

So is there any helpful information you can give me e.g. how do you adjust for lot to lot variations?

Another question I have is: what isotype control do you suggest people use when the antibody is a 'tissue culture supernatant' with unknown IgX conc ?

Answer

Thank you for your enquiry.

I can confirm that many of our antibodies are supplied in purified format, and we can provide the concentration forthese.

However, unpurified antibodies, such as those sold as whole antiserum, ascites or tissue culture supernatant will not have a concentration stated on the datasheet. Antibody concentration is usually determined by protein assay, and serum / ascites / tissue culture supernatant will contain a lot of other proteins, which means the antibody quantification would not be accurate. The concentration is therefore not determined.

As far as I am aware, this situation would be the same for any company selling antibodies.

I am able to provide an estimate. Generally, for tissue culture supernatant antibodies, concentration of antibody is known to vary between 1 - 3 mg/ml.

I do understand that an estimate does make this tricky when selecting a concentration to use the isotype control, however I hope this will help. Regrettably we do not currently have a rabbit monoclonal IgG isotype control supplied in TC supernatant format.

For future reference:
For whole antiserum, concentration of antibody is known to very between 1 - 10 mg/ml.
For ascites, concentration of antibody is known to very between 5 - 10 mg/ml.
