    NF-kappa-B is a pleiotropic transcription factor which is present in almost all cell types and is involved in many biological processed such as inflammation, immunity, differentiation, cell growth, tumorigenesis and apoptosis. NF-kappa-B is a homo- or heterodimeric complex formed by the Rel-like domain-containing proteins RELA/p65, RELB, NFKB1/p105, NFKB1/p50, REL and NFKB2/p52 and the heterodimeric p65-p50 complex appears to be most abundant one. The dimers bind at kappa-B sites in the DNA of their target genes and the individual dimers have distinct preferences for different kappa-B sites that they can bind with distinguishable affinity and specificity. Different dimer combinations act as transcriptional activators or repressors, respectively. NF-kappa-B is controlled by various mechanisms of post-translational modification and subcellular compartmentalization as well as by interactions with other cofactors or corepressors. NF-kappa-B complexes are held in the cytoplasm in an inactive state complexed with members of the NF-kappa-B inhibitor (I-kappa-B) family. In a conventional activation pathway, I-kappa-B is phosphorylated by I-kappa-B kinases (IKKs) in response to different activators, subsequently degraded thus liberating the active NF-kappa-B complex which translocates to the nucleus. NF-kappa-B heterodimeric p65-p50 and p65-c-Rel complexes are transcriptional activators. The NF-kappa-B p65-p65 complex appears to be involved in invasin-mediated activation of IL-8 expression. The inhibitory effect of I-kappa-B upon NF-kappa-B the cytoplasm is exerted primarily through the interaction with p65. p65 shows a weak DNA-binding site which could contribute directly to DNA binding in the NF-kappa-B complex. Associates with chromatin at the NF-kappa-B promoter region via association with DDX1.

    Monomethylated at Lys-310 by SETD6. Monomethylation at Lys-310 is recognized by the ANK repeats of EHMT1 and promotes the formation of repressed chromatin at target genes, leading to down-regulation of NF-kappa-B transcription factor activity. Phosphorylation at Ser-311 disrupts the interaction with EHMT1 without preventing monomethylation at Lys-310 and relieves the repression of target genes.
    Phosphorylation at Ser-311 disrupts the interaction with EHMT1 and promotes transcription factor activity (By similarity). Phosphorylation on Ser-536 stimulates acetylation on Lys-310 and interaction with CBP; the phosphorylated and acetylated forms show enhanced transcriptional activity.
    Reversibly acetylated; the acetylation seems to be mediated by CBP, the deacetylation by HDAC3. Acetylation at Lys-122 enhances DNA binding and impairs association with NFKBIA. Acetylation at Lys-310 is required for full transcriptional activity in the absence of effects on DNA binding and NFKBIA association. Acetylation can also lower DNA-binding and results in nuclear export. Interaction with BRMS1 promotes deacetylation of 'Lys-310'.

      Western Blot with ab19870 after p65 Immunoprecipitation: rabbit polyclonal to NF-kB p65 (acetyl K310) (ab19870; 2.5µg/ml) in 1% non-fat milk TBS-T incubated for 3 hours at room temperature. Exposure time: 1 min normal ECL. Tested samples: nuclear extracts (180 µg) of immortalized p65-/- mouse cells, complemented with the empty vector (pRRL), wild-type p65 (Wt) and non-acetylatable K310 (K310R). The samples tested were treated with deacetylase inhibitors HDACi (TSA + Nicotinamide) and TNF-alpha. The samples were immunoprecipitated with 2µg of alpha-p65 and subsequently analysed by Western blot with Rabbit polyclonal to NF-kB p65 (acetyl K310) (ab19870).       ab19870 recognizes Rabbit polyclonal to NF-kB p65 (acetyl K310) specifically at ~75kDa (indicated by the arrow) is this SDS-PAGE blot. The p65 band runs higher than 65kDa in this SDS-PAGE blot as it contains a myc-tag. We are sure that the band at ~75kDa is p65 since p65 specific antibodies detect the same band in IP and WB and there is no signal in the p65 knock-out cell line with ab19870. A number of additonal bands are recognized by ab19870 when tested with endogenous p65 from whole cell extracts, we do not know the identity of these bands.

    Inquiry: Dear Sir/madam, I purchased one of your antibody products, anti NFkB p65 (ab19870). I used the antibody to make CO-IP from my nuclear K562 cell lysate. I used a commercial CO-IP kit with magnetic beads. After CO-IP i made western blot to check out interacting proteins. I incubated membrane with the same antibody to see if i successfully precipitated my protein of interest (p65). My problems are: After incubation with NFkB p65 and proper secondary antibody, i saw 3 bands on the membrane: One of them (the strongest signal) is nearly 55 kDa (according to the marker), the other one which is much weaker is below 25 kDa and the last and very fainted one is bigger than 90 kDa. I could not understand why i saw three bands, because after CO-IP i should have a purified protein sample. If i consider the band near 55 kDa as NFkB p65 (because its signal is really very strong) I also could not understand why i did not see my protein band at 65 kDa, is it possible to see NFkB p65 protein at 55 kDa (since i loaded the protein lysate with magnetic beads can it have an effect on migration?)?     Question

    I've attached my western blots with this email. I tried your antibody twice.
    1) Primary for 2.5ug/ml for 3hrs (according to protocol), secondary: 1:5000 (anti-rabbit)
    2) Primary for 1:1000 overnight, secondary: 1:5000 (anti-rabbit)

    Lot number is GR63202-2. Not sure about the order number. Where can I find that?

    I did not perform an IP prior because it is impractical to collect the number of limb samples required for this procedure.

    There limbs samples are from mice (GD12 CD1 pregnant). RIPA lysis buffer used. HDAC inhibitor (valproic acid) is administered during limb cultures in BGJB/salt solution for specific time points. The samples I used here is the 12hr incubation time-point.

    Samples are boiled @ 95oC for 5mins. Loading buffer contains B-mercaptoethanol. Each well contains 20ug of protein. Blocking buffer is 5% non-fat milk in TBST for 1hr before primary incubation.

    Hello,
    I would like to test the Anti-NFkB p65 (acetyl K310) antibody -
    ChIP Grade (ab19870) in IHC of the same paraffin-embedded mouse prostate
    tissue. According to the datasheet, this antibody recognizes the mouse
    protein, but it hasn't been tested for IHC. Could you let me know if I
    would get the 100% testing discount if I do the test and submit the
    abreview?
    Also, I welcome any suggestions for the tests that might not be included
    in the datasheets. Or suggestions of other antibodies that might be more
    suitable for my needs.
