    Neuro-2A cells stained for Mu Opioid Receptor (green) using ab17934 at 1/250 dilution in ICC/IF. It was followed by Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate at 1/2000 dilution for 45 minutes at room temperature (Panel a). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI. F-actin (Panel c: red) was stained with Rhodamine Phalloidin at 1/300 dilution. Panel d represents the merged image showing membranous localization. Panel e shows the no primary antibody control.

Estoy interesada en un anticuerpo anti-receptor mu-opioide de rata que reconozca un epitope extracelular. He encontrado dos anticuerpos en el sitio web que reconocen el extremo N-terminal, pero del receptor humano, y hay otros dos, que reconocen rata, pero cuyos inmunógenos son péptidos sintéticos de os cuales no se aclara exactamente la posición.

Después de consultar este caso con mis compañeros del laboratorio, acerca de los siguientes anticuerpos contra Mu Opioid Receptor validados en tejidos de rata: ab10275, ab17934, ab64746, ab128013 y ab134054, me temo que ninguno de ellos ha demostrado reconocer el epítopo extracelular de la proteína.
Todos ellos se han generado a partir del extremo C terminal de la proteína, por lo que, a pesar de no haberse mapeado el epítopo que reconocen, probablemente detecten el dominio citoplásmico de la misma.
La solución que te propongo es testar uno de los anticuerpos que hay en el catálogo hecho a partir del extremo N terminal de la proteína, y testarlo en rata. Tenemos en abcam un sistema de evaluación de anticuerpos por el cual al testar uno de ellos en una aplicación o especie que no se ha testado, puedes beneficiarte de un descuento en tu próxima compra por valor del anticuerpo, si lo usas en esa especie o aplicación nueva y nos mandas los resultados a la web en forma de Abreview.

Question

We have tried using this receptor antibody, along with Rab5 to investigate Mu opioid receptor trafficking in rat brain tissue sections.
The rats have been treated with either saline or two compounds dissolved in saline by IP injection.
The brains have then been removed and snap frozen using isopentane and stored in the -80deg freezer before sectioned in a crystat to 20um thick.
We have tried optimizing fixation (PFA, acetone, methanol), permeableisation (triton-x, tween, saponin), antibody concentration (primary and secondary) and duration/temperature, blocking (times and concentration of blocking).
We have ascertained good staining in thalamus where Mu opioid receptors are abundantly expressed on interneurons and cortex where mu expressed, but to a lesser degree to that in thalamus.
However, we have only observed green (mu secondary = fluoro488) staining consistent with astrocytic/glail neurons, no interneurons or pyramidal neurons are stained.
We would expect little or no astroglial mu opioid receptors in thalamus or cortex.
We were wondering if you might be able to shed some light on what this staining might be specific to on these cells and how we might go about getting some specific mu opioid receptor staining in actually neurons.

Question

Problem: Antibody not working in WB Primary was used at 1:1000 and 1:500 dilutions, overnight incubation at +4C, in 5% skim milk in TBST (tween 0.1%). The secondary antibody is a donkey anti-rabbit HRP conjugated from GE Healthcare used at 1:20000, routinely used in our lab. The samples are membranes from CHO cells transfected with rat MOR-1 and one splice variant of it that has also the same epitope (both should be recognized, but none is ). Samples from mock tranfected CHOs, rat MOR-1 transfected CHOs and rat MOR-1 variant transfected CHOs were probed in parallel also with another rabbit polyclonal (this one working fine) that is showing that our receptors are indeed expressed. The gel was run in non reducing conditions, membranes were extracted in a buffer containing 2mM DTT.

Although your samples are showing receptor expressio with another product, Abcam antibodies will have been tested under reduced and denatured conditions only and we recommend using reducing and denaturing conditions unless otherwise stated on the datasheet. Thank you for your enquiry and for taking your time to fill in the on-line Questionnaire. As we understand from your e-mail, the customer used mouse Mu Opioid Receptor expressing HEK cells. We would like to emphasize that this antibody was rtaised against synthetic peptide of human Mu Opioid Receptor. Therefore the antibody recognizes human protein, but unfortunately other species i.e. mouse has not been tested for cross-reaction. Although, there is a chance that it may work in this species due to 94% sequence homology. 1. We would suggest running a good positive control along with the samples, lysate from human brain or spinal cord would be a good candidate. 2. We do not have information about the Bock Ace, so would rather suggest using 5% BSA-TBST for blocking. 3. Please make sure that the customer use RIPA buffer for sample preparation: RIPA Base Ingredients Tris-HCl: 50 mM, pH 7.4 NP-40: 1% Na-deoxycholate: 0.25% NaCl: 150 mM EDTA: 1 mM PMSF: 1 mM Aprotinin, leupeptin, pepstatin: 1 microgram/ml each RIPA Protease Inhibitors Phenylmethylsulfonyl fluoride (PMSF) (200 mM stock solution in isopropanol; store at room temperature) EDTA (calcium chelator; 100 mM stock solution in H2O, pH 7.4) Leupeptin (store frozen in aliquots, 1 mg/ml in H2O) Aprotinin (store frozen in aliquots, 1 mg/ml in H2O) Pepstatin (store frozen in aliquots, 1 mg/ml in methanol) We hope this information will be useful.
