Mouse Neurofilament H ELISA kit (ab272479) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Mouse Neurofilament H protein in tissue culture extract. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse Neurofilament H with 57.12 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. -Single-wash protocol reduces assay time to 90 minutes or less

特記事項
Neurofilament Heavy Polypeptide (also known as Neurofilament H or NEFH), together with Neurofilament L and M are three intermediate filament proteins of neurofilaments. Neurofilament H is involved in the maintenance of neuronal caliber and may play a role in intracellular transport. NEFH is used as a neuronal damage biomarker and mutations to the gene have been associated with susceptibility to amyotrophic lateral sclerosis. The mouse NEFH amino acid sequence is 94.4% and 96.9% homologous with the human and rat NEFH sequence, respectively.

Neurofilaments usually contain three intermediate filament proteins: L, M, and H which are involved in the maintenance of neuronal caliber. NF-H has an important function in mature axons that is not subserved by the two smaller NF proteins. -

Defects in NEFH are a cause of susceptibility to amyotrophic lateral sclerosis (ALS) [MIM:105400]. ALS is a neurodegenerative disorder affecting upper and lower motor neurons, and resulting in fatal paralysis. Sensory abnormalities are absent. Death usually occurs within 2 to 5 years. The etiology is likely to be multifactorial, involving both genetic and environmental factors. -

There are a number of repeats of the tripeptide K-S-P, NFH is phosphorylated on a number of the serines in this motif. It is thought that phosphorylation of NFH results in the formation of interfilament cross bridges that are important in the maintenance of axonal caliber.
Phosphorylation seems to play a major role in the functioning of the larger neurofilament polypeptides (NF-M and NF-H), the levels of phosphorylation being altered developmentally and coincident with a change in the neurofilament function.

Interpolated concentrations of native Neurofilament Heavy Polypeptide in mouse brain tissue extract based on a 12.5 µg/mL extract load.
The concentrations of Neurofilament Heavy Polypeptide were measured in duplicate and interpolated from the Neurofilament Heavy Polypeptide standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Neurofilament Heavy Polypeptide concentration was determined to be 5491.67 pg/mL in brain extract.
