Mouse MASP1 ELISA kit (ab272473) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Mouse MASP1 protein in serum and plasma. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse MASP1 with 29.3 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 特記事項
MASP1, encoded by the Masp1 gene, is a serine protease and the most abundant protease of the lectin pathway. MASP1 plays a central role in complement activation. The binding of mannose binding lectin (MBL) and ficolins to sugar moieties triggers the lectin pathway and leads to the activation of MASP1 that will recognize and neutralize pathogens through patterns of sugar moieties. Studies have shown MASP1 to promote coagulation and take an active role in the inflammation process. Human, rat, cow, and monkey share 90%, 97%, 89%, and 90% sequence homology, respectively.

MASP is a component of the bactericidal ra-reactive factor rarf which specifically binds to RA and R2 polysaccharides expressed by certain enterobacteria. It triggers the activation of complement cascade by activating the C4 and C2 components. It activates the C4 component by cleaving the alpha-chain of C4. The alternative-splicing product from MASP1 gene is called MASP3. MASP1 is associated with smaller MBL oligomers whereas MASP3 is found on larger oligomers. The substrate of MASP3 is unknown. -

The MASP1 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of MASP1 were measured in duplicates, interpolated from the MASP1 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1:400, plasma (citrate) 1:400, and plasma (EDTA) 1:200. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MASP1 concentration was determined to be 902.1 ng/mL in serum, 416.2 ng/mL in plasma (citrate) and 779.9 ng/mL in plasma (EDTA).
