    Abcam’s HGF Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse HGF in serum, plasma and cell culture supernatants.

    This assay employs an antibody specific for mouse HGF coated on a 96- well plate. Standards and samples are pipetted into the wells and HGF present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse HGF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of HGF bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

The antibody pair inab100686 recognizes an epitope located somewhere between aa 33-728 (which is whole sequence minus the signal peptide). Therefore the kit is expected to detect the pro (uncleaved) form, the heterodimeric form, the alpha subunit, and the beta subunit (so total HGF). Please note that we have not specifically tested each form separately but both antibodies are polyclonal so it is likely.

I've heard back from my contact at the lab, and the standard is biologically active. Activity was measured as the ability to stimulate H-thymidine incorporation in 4MBr-5 monkey cells. The ED50 is typically 20-40 ng/mL.

Thank you for contacting Abcam. Unfortunately, epitope mapping has not been performed for this kit so we do not know exactly where the antibodies are binding.  Therefore whether the kit will recognize HGF bound to the c-met receptor or inhibit c-met from binding, is not known.   I wish I could be of more help on this one. The best kit to use for tissue samples is ab100687 HGF Mouse ELISA Kit - 1 x 96 Well Plate. This is specifically meant for tissue samples. Both of the antibodies in the kit are polyclonal and considering that the homology between mouse and rat is high (around 98%), the odds are high that these kits would be suitable for rat samples. However, we have not tested this and are unable to give guarantee.
