HeLa cells were fixed with 4% paraformaldehyde in PBS for 10 minutes, followed by blocking/permeabilization in 5% Normal Goat Serum/0.3% Triton X-100 in PBS for 1 hour. Coverslips were washed and incubated with primary antibody (1:200) for 1 hour, washed, and incubated with secondary antibody (1:1000) for 45 minutes.
The image shown is a maximum intensity projection of a z-stack taken on a spinning disk confocal microscope. The antibody stains the eadosomal/lysosomal compartment as expected.
