The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
1/200. Predicted molecular weight: 58 kDa.
Use at 20 µg/mg of lysate.
Use a concentration of 5 µg/ml.
ab171870 - Rabbit polyclonal IgG, is suitable for use as an isotype control with this antibody.
Use a concentration of 1 µg/ml.
Use a concentration of 10 µg/ml.
Histone demethylase that specifically demethylates 'Lys-9' of histone H3, thereby playing a central role in histone code. Does not demethylate histone H3 'Lys-4', H3 'Lys-27', H3 'Lys-36' nor H4 'Lys-20'. Demethylates both di- and trimethylated H3 'Lys-9' residue, while it has no activity on monomethylated residues. Demethylation of Lys residue generates formaldehyde and succinate.
Belongs to the JHDM3 histone demethylase family. Contains 1 JmjC domain. Contains 1 JmjN domain.
ab93694, at 10µg/ml, staining KDM4D / JMJD2D in DLD1 (Human Colorectal Adenocarcinoma) cells by Immunofluorescence. Cells were fixed with 1% formaldehyde, permeabilized with 0.1% Triton X-100 and blocked with normal goat serum. Secondary antibody: Goat anti Rabbit Cy3 at 1/250 dilution.
IHC image of ab93694 staining in Human breast cancer formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab93694, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
