      Ubiquitin-like protein that is conjugated to intracellular target proteins after IFN-alpha or IFN-beta stimulation. Its enzymatic pathway is partially distinct from that of ubiquitin, differing in substrate specificity and interaction with ligating enzymes. ISG15 conjugation pathway uses a dedicated E1 enzyme, but seems to converge with the Ub conjugation pathway at the level of a specific E2 enzyme. Targets include STAT1, SERPINA3G/SPI2A, JAK1, MAPK3/ERK1, PLCG1, EIF2AK2/PKR, MX1/MxA, and RIG-1. Deconjugated by USP18/UBP43. Shows specific chemotactic activity towards neutrophils and activates them to induce release of eosinophil chemotactic factors. May serve as a trans-acting binding factor directing the association of ligated target proteins to intermediate filaments. May also be involved in autocrine, paracrine and endocrine mechanisms, as in cell-to-cell signaling, possibly partly by inducing IFN-gamma secretion by monocytes and macrophages. Seems to display antiviral activity during viral infections.
      In response to IFN-tau secreted by the conceptus, may ligate to and regulate proteins involved in the release of prostaglandin F2-alpha (PGF), and thus prevent lysis of the corpus luteum and maintain the pregnancy.

      Bill, Thanks for looking into my CAPG Ab. I will let you know what I find out. I have another technical question for you. I am also using ab14374, rabbit pAb to ISG15. The notes state that with WB, a band of about 18.5 kDa is recognized and the Ab may recognize other prominent intrinsic bands. In my hands, I see a prominent band of the correct size even at 1:500 dilution and I also see several "prominent intrinsic bands" 30-150 kDa in size. Do you know of anyone who used this Ab that DID NOT see these extra bands? The Malakhov paper sees the same thing I do. I would also like to use this antibody for ELISA and it is indicated in the Notes that this has been attempted. I am afraid that such prominent extraneous bands will lead to a false positive with ELISA. Do you have any information as to whether this is one antibody recognizing several other proteins or if there are other concentrated antibodies in the Ab solution? If it is the latter, an Ab purification should do the trick. Please let me know your thoughts. Thank you,

      Thank you for your enquiry. No, I do not know of anyone that did not see these extra bands. It is expected to see these bands. My understanding is that these extra bands come from the binding of other proteins to ISG15 and that these proteins are not separated from ISG15 during the sample preparation and running on an SDS-PAGE gel because they are coupled to ISG15. This is one antibody recognizing ISG15. I would not expect an antibody purification to change your result. The antibody will detect ISG15 in an ELISA, however, it may also detect ISG15 that is covalently linked to other proteins, conjugates, or ubiquitin-like modifiers.       The datasheets for these two antibodies are exactly the same. The only difference is that ab14374 is listed as 500 ug and ab14687 as 250 ug.
