      Isoform 2 is a receptor for IL22. Binds to IL22, prevents interaction with the functional IL-22R complex and blocks the activity of IL22 (in vitro). May play an important role as an IL22 antagonist in the regulation of inflammatory responses.
      Isoform 1 may play a role in establishing and maintaining successful pregnancy.

      Expressed in placenta, spleen, breast, skin and lung. Also detected in intestinal tract, testis, brain, heart and thymus. No expression found in prostate, bladder, kidney, ovary, muscle, bone marrow, liver and uterus. Isoform 1 is expressed only in placenta. Isoform 2 is expressed in placenta and breast and at lower level in spleen, skin, thymus and stomach.

    ich habe vor einiger Zeit mit Ihnen telefonisch Kontakt aufgenommen und per email einen Überblick über mein Versuchsprotokoll an Sie weitergeleitet, da ich trotz zahlreicher Versuche die Immunhistochemie mit dem oben genannten Antikörper gegen IL-22BP nicht reproduzieren konnte. Als Positivkontrollen wurden wie im data sheet humane Milzproben wie auch Tonsillenproben genutzt. Sie schlugen mir vor, nochmals mit dem Testlabor Rücksprache zu halten, welches Protokoll von Abcam genutzt wurde, um die im data sheet abgebildeten Färbungen von IL-22BP zu ermöglichen.



    Ich frage Sie nun, ob Sie nähere Informationen vom Labor erhalten haben oder sich andere Anhaltspunkte beim Abgleichen meines Protokolles bzw der Lot No ergeben haben.

    Es tut mir Leid für diese Verzögerung und das Sie Probleme mit unserem Antikörper haben. Ich habe leider noch keine Bestätigung aus dem Labor, bezüglich des verwendeten Protokolls erhalten.

    Ihre mir zugesendeten Protokollinformationen sind sehr hilfreich für unsere Qualitätskontrolle und ich möchte Ihnen noch einmal Danken, für die Zeit die Sie sich genommen haben unseren Fragebogen auszufüllen.

    Ich verstehe, dass Sie wegen der schlechten Resultate, die Sie mit diesem Antikörper erzielt haben beunruhigt sind. Ich weiß sehr zu schätzen wie viel Zeit Sie schon im Labor verbracht haben bei dem Versuch das Ergebnis zu verbessern.

    Wie ich an Hand ihres Protokolls sehen kann haben sie gründlich gearbeitet und es gibt nicht viele Verbesserungsvorschläge die ich machen kann. Ich bedauere, dass Sie wahrscheinlich ein schlechtes Röhrchen erhalten haben.

    Ich entschuldige mich für die Umstände und würde Ihnen gerne als Kompensation ein Ersatzprodukt oder eine Gutschrift anbieten. Ich werde dem Labor noch einmal eine Errinerungsemail schreiben und sobald ich nähere Protokollinformationen aus dem Labor erhalte werde ich diese an Sie weiter leiten.

    Die einzigen Vorschläge, die ich gerne zu Ihrem Protokoll machen würde sind folgende:

    1. In unseren Standardprotokollen empfehlen wir mit 4% formalin zu fixieren. Je höher konzentriert und je Länger fixiert wird, desto mehr können Antigene maskiert werden.

    2. Es könnte hilfreich sein den HIER Antigen retrieval über einen längeren Zeitraum (bis 20 min) auszuführen.

    Es tut mir leid, dass ich Ihnen im Moment nicht mehr weiterhelfen kann. Ich freu mich von Ihnen zu hören, welche Alternative Sie als Kompensation bevorzugen.
