    Abcam’s Thrombomodulin (CD141) Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Thrombomodulin (CD141) in serum, plasma, buffered solutions and supernatants.

    A monoclonal antibody specific for Thrombomodulin has been coated onto the wells of the microtiter strips provided. Samples, including standards of known Thrombomodulin concentrations, control specimens or unknowns are pipetted into these wells. During the first incubation, the standards or samples and a biotinylated monoclonal antibody specific for Thrombomodulin are simultaneously incubated. After washing, the enzyme Streptavidin-HRP, that binds the biotinylated antibody is added, incubated and washed. A TMB substrate solution is added which acts on the bound enzyme to induce a colored reaction product. The intensity of this colored product is directly proportional to the concentration of Thrombomodulin present in the samples.

    This kit will recognize both endogenous and recombinant Human Thrombomodulin.

    Thrombomodulin is a specific endothelial cell receptor that forms a 1:1 stoichiometric complex with thrombin. This complex is responsible for the conversion of protein C to the activated protein C (protein Ca). Once evolved, protein Ca scissions the activated cofactors of the coagulation mechanism, factor Va and factor VIIIa, and thereby reduces the amount of thrombin generated.

    Defects in THBD are the cause of thrombophilia due to thrombomodulin defect (THR-THBD) [MIM:188040]. A hemostatic disorder characterized by a tendency to thrombosis.
    Defects in THBD are a cause of susceptibility to hemolytic uremic syndrome atypical type 6 (AHUS6) [MIM:612926]. An atypical form of hemolytic uremic syndrome. It is a complex genetic disease characterized by microangiopathic hemolytic anemia, thrombocytopenia, renal failure and absence of episodes of enterocolitis and diarrhea. In contrast to typical hemolytic uremic syndrome, atypical forms have a poorer prognosis, with higher death rates and frequent progression to end-stage renal disease. Note=Susceptibility to the development of atypical hemolytic uremic syndrome can be conferred by mutations in various components of or regulatory factors in the complement cascade system. Other genes may play a role in modifying the phenotype.

Answer

Thank you very much for your understanding. The things are more clearer now. Lab has confirmed that the number K100624 (typos K10624) is indeed stands for standard vial.

They have also confirmed that the standard conc. should be between 11-16 ng/ml at 1ml reconstitution with diluent. The expected range value 11-16 ng/ml has been determined by 2 operators, 3 experiments, 6 replicates each.

So at this moment I am unsure about 7ng/ml reading of standard, which is about ˜50% lower than the expected. To understand this I have few questions to ask;

- Does all the 5 kits show conc. range of 7ng/ml?
- I have checked the order history of this kit and found that the same item was bought many times. Would you be so kind, letting me know the lot numbers affected by this problem. You have received >5 kits of batch number http://it.adminsite.com/admin/stock/stock_list.cfm?intAbID=46508&intStockID=1119463&fMain=1&fViewExpired=1&strViewOffice=%25 was this batch fine?
- Do you follow the protocol exactly as given on the datasheet? Have you changed any step?
- From you r experience do you think, only the standard is problematic? If you think other can also be, then which one you would choose?

My colleague is out of office and he has asked me to look after her customers during her absence.


The whole Thrombomodulin kit is stable if refrigerate (4°C) until at least for one year. However, we would suggest our customers to use it as soon as possible after purchase to make sure that our Abpromise guarantee applies (6 months after purchase).

The cytokine provided (Thrombomodulin lyophilized standard vial) is stable at 4°C several years. When the material is reconstituted, the stability is not guarantee even if frozen at -70°C. The vial should be resuspended immediately prior to use and then discard.
