Please recommend the customer to mix few micro litres of TMB solution with HRP-streptavidin solution on a glass slide. The colour developed should be blue which will change to yellow when stop solution is added.

The problem may also be due to sample being not having Procalcitonin or the customer might be using very diluted samples. Please suggest customer to check the dilutions etc.

1A, For storage please refer to the protocol and store the individual as given in the booklet.

"ab100630 may be stored for up to 6 months at 2 to 8°C from the date of shipment. Standard (recombinant protein) should be stored at -20°C or -80°C (recommended at –80°C) after reconstitution. Opened Microplate Wells or reagents may be store for up to 1 month at 2 to 8°C. Return unused wells to the pouch containing desiccant pack, reseal along entire edge. Note: the kit can be used within one year if the whole kit is stored at -20°C. Avoid repeated freeze-thaw cycles"

The kit is guaranteed for 6 months from he data of purchase if the components are stored correctly as given in booklet.

2A: Generally we recommend diluting plasma and serum to 1/10 or 1/50 dilution. The correct dilution factor however should be determined by end user.

B; There are two vial of detection antibody in kit. If you add 100ul of assay buffer in each vial you will get 200ul concentrated antibody. Now dilute it further 80 times as recommended in step 6 then the total volume will be 16ml. Now at 100ul per well you will only need 9.6 ml for 96 well plate. The volume you will left is 6.4 ml which is more than enough.

The micro well comes in 12 strips of 8 wells. You cannot break 1 strip of 8 wells further. It is possible but may not be easy.

I do not understand 1/2 well. Please clarify.

The 8 well strips provided with ab100630 are not designed for single wells to be broken off and used individually.

I am sorry for the delay in getting back to you. We are not open on the weekends. I am now able to confirm that you should be able to isolate individual wells of this product. As discussed in my previous email, the plates are provided as 12 strips of 8 wells each. You should be able to divide these further by using a scalpel or scissors.

The microplate provided with the Procalcitonin Human ELISA Kit (ab100630) is composed of 12 strips of 8 wells. This allows for experiments to be performed at different times simply by taking as many wells as is required and storing the rest.

In answer to your second question, we do not have set expiration dates for our products. We strongly recommend that you follow the storage instructions on the datasheet and protocol for the product you purchased. These conditions will vary among our products, therefore, it is important to verify the storage conditions for each of our products when you receive them. We guarantee all of our products to work for at least 120 days from the date of purchase when stored correctly.

these has not been tested with Swine or pig samples so we are unable to confirm the species cross reactivity. It mean only ab100755, ab100756, ab108674, ab108794, ab113352, ab113353 are OK to use with Porcine samples and are guaranteed. Would you like to have a quote for these antibodies.

The Procalcitonin Human ELISA Kit (ab100630) has to our knowledge not been compared to performing the Brahms method of detecting human procalcitonin. The kit should however match up well with most of the reported literature which says that normal levels of procalcitonin in serum and plasma are in the low pg/ml range.

Your customer should however be made aware that most of the normal serum and plasma samples we have measured using our kit showed procalcitonin levels were below the sensitivity limit of the kit, which is 10 pg/ml. Procalcitonin levels being below the detection limit seems to be very common among procalcitonin ELISA kits in general so testing samples which have expected elevated levels of procalcitonin would likely to be more applicable.
