    Human IL-1 R4 ELISA Kit (ST2) (ab100563) is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human IL-1 R4 in serum, plasma, and cell culture supernatants.


    This assay employs an antibody specific for Human IL-1 R4 coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-1 R4 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human IL-1 R4 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-1 R4 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

    ST2 is a member of a superfamily containing the Interleukin 1 (IL1) receptor and the Toll-like receptors (TLRs). The TLRs are signaling molecules that recognize different microbial products during infection and serve as an important link between the innate and adaptive immune responses. ST2 was originally identified as a protein whose production was stimulated by various proliferation-inducing agents such as PDGF and FGF. More recently, it has been shown to negatively regulate IL1 receptor and Toll-like receptor 4 (TLR4) signaling and to maintain endotoxin tolerance. It has been suggested that the inhibition of TLR4 signaling occurs through the association and sequestering of TLR adaptor molecules such as MyD88 and TIRAP.

    Inquiry: I would clarify for myself one thing with human IL-1 R4 ELISA kit. Detection range is 2 to 1200 pg/ml, and recommended dilution for serum and plasma is 2-fold. However reviewing papers on ST2 level in patients with cardiovascular diseases (stable, no advanced heart failure) I found the usual range is between approx. 10 and 50 ng/ml while in self-reported healthy persons it may reach approx. 30 ng/ml. So dilution should be 50-fold to get into detection range. One the contrary there are only few papers, which give ST2 levels within approx. 10-200 pg/ml (i.e. within Abcam detection range), and only after major cardiac surgery like as CABG temporal increase up to approx. 15 ng/ml occurs. What is this difference in units and levels driven by? I do understand I have to derive our own dilution factor, but at preanalytical stage I'm a bit confused with previous studies.

    The detection range for ab100563 Human IL-1 R4 ELISA Kit (ST2) is based on the standard curve range and from testing of normal human serum with the kit.  Levels of IL-1 R4 in these samples using this kit were about 150 pg/ml).

    Each kit manufacturer will use different antibodies and standards i their kits. This can result in slightly different detection ranges and normal levels.  Dilution factors will need to be individually optimized for the individual samples by customers.  The recommended dilution provided is from our testing of normal human serum and plasma.

    J’ai contacté le laboratoire concernant le kit ab100563. L'immunogène utilisé pour créer les anticorps est une protéine recombinant humain IL-1R4 qui correspond à la région extracellulaire (acides aminés19-328). Malheureusement les épitopes de ces anticorps ne sont pas déterminés. J’ai donc comparé la région extracellulaire avec la séquence de la protéine du rat. L’homologie totale est de 62% mais il y’a des régions très bien conservé et d’autres qui ne sont pas du tout conservé. Je peux quand même vous faire l’offre de test mais il est très difficile de prédire si ce kit fonctionnera chez le rat.
