    Abcam’s ICAM1 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human soluble ICAM1 in serum, plasma and cell culture supernatants.


    This assay employs an antibody specific for Human ICAM1 coated on a 96-well plate. Standards and samples are pipetted into the wells and ICAM1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human ICAM1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of ICAM1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

    ICAM1 is a 85-110 kDa single chain type 1 integral membrane glycoprotein with an extracellular domain of five immunoglobulin superfamily repeats, a transmembrane region and a cytoplasmic domain. It shares considerable amino acid sequence homology with ICAM3 and with ICAM2. ICAM1 is expressed by activated endothelial cells. It is detected on cells of many other lineages (e.g. epithelial cells, fibroblasts, chondrocytes, B lymphocytes, T lymphocytes (low), monocytes, macrophages, dendritic cells and neutrophils), with lower levels that increase in inflammation. ICAM1 is also detected in some carcinoma and melanoma cells. Soluble ICAM1 is detectable in the plasma and is elevated in patients with various inflammatory syndromes. It is the receptor for rhinoviruses and malaria.

Here is the requested information: ab46087: Normal values for TNFa in such samples are negatives or very low, so we usually recommend to assay the human serum samples neat with Standard buffer, human serum to dilute the standard curve, or at the 1:2 dilution with standard buffer diluent PBS BSA (in that case, the standard curve should be diluted in same buffer). Below are our recommended normal human serum dilutions for the other 3 kits. These recommendations should only be used as a starting point and the optimal sample dilution needs to be determined by the enduser empirically based on researched literature and knowledge of their particular sample. ab100586: 2 fold dilution ab100610: 500-2000 fold dilution ab100640: 5-200 fold dilution I hope this information is helpful to you.     Yes the ICAM in human serum/plasma is in the 200-300 ng/ml range but the high end of the curve for the Abcam kit (AB100640) is 2000pg/ml or 2 ng/ml. So do we need a 200-300 fold dilution before assaying. That was the question we posed

    I purchased the following kits and there are issues with the sample range: ab100640, ICAM1 Human ELISA Kit - 1 x 96 - Normal human sample range is from 0.2-1pg/ml. However the kits range is 24.58 pg/ml - 6000 pg/ml.  Will the kit recognises samples as low as normal human range?

    Thank you for contacting Abcam. I have received a reply from the lab about ab100640, the ICAM1 Human ELISA Kit. Based on testing and the reported literature, normal levels of soluble ICAM in serum look to be in the low-to-mid ng/ml levels and not low pg/ml levels. See below for a few references.
