      Abcam’s anti-Herpes simplex virus Type 1 (HSV1) IgG Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate qualitative measurement of IgG class antibodies against Herpes simplex virus Type 1 in Human serum and plasma.

      A 96-well plate has been precoated with Herpes simplex virus Type 1 antigens to bind cognate antibodies. Controls or test samples are added to the wells and incubated. Following washing, a horseradish peroxidase (HRP) labelled anti-Human IgG conjugate is added to the wells, which binds to the immobilized Herpes simplex virus Type 1-specific antibodies. TMB is then catalyzed by the HRP to produce a blue color product that changes to yellow after adding an acidic stop solution. The density of yellow coloration is directly proportional to the amount of Herpes simplex virus Type 1 IgG sample captured in plate.

      Serologic ELISA assay principle
      Specific antigens are coated on the 96-well plate, controls or test samples are added to the well and incubated. The wells are washed to remove any unbound Human anti-antigen antibodies (Ig). A horseradish peroxidase (HRP) labelled anti-Human Ig conjugate is added to the wells. TMB is then catalyzed by the HRP to produce a blue color product that changes to yellow after adding an acidic stop solution. The intensity of yellow coloration is directly proportional to the amount of Human anti-antigen Ig captured on the plate.

    The lab sent the following advice:

    "The HSV-1 IgG ELISA Kit is a qualitative immunoassay to determine the presence or absence of IgG to HSV-1. The end result is not quantitative, however, the relative result is indicative of higher or lower titer.
    The researcher can compare the results of different test runs to infer relative titers. The researcher should be catious, however, that they are not measuring run to run variability or even a slight decline in the readings as the kit ages.
    Assuming the researcher is testing samples taken from different time points, it may be best to test the samples side by side within the same run as they are collected for relative comparison of antibody titer."

    It might be advisable - if that is possible in your experimental set up - to collect all the samples and then run the assay for all samples together at the end (e.g. after 6 months) to be able to compare them.

    I have obtained the following additional information:

    The kit can be used for semi-quantification. This means that the result in OD can be expressed in NTU (arbitrary units). Therefore the OD of the sample is divided by the OD of the cut-off. This result is multiplied by 10. The result in NTU can be compared between different samples and different test runs. It is described in section 9 of the protocol

    It is not recommended to use the positive control for a standard curve since the linearity is not determined.

    In general the concentration of the positive control is not determined in detail. For a qualitative test it is sufficient that the positive control is higher than the cut-off.

    I received the following information from the lab:

    The HSV-1 IgG ELISA kit (ab108737) is intended for the qualitative determination of IgG to HSV-1. The kit is not designed to provide a quantitative result.

    The lab is looking intothe concentration of the positive control and will let me know.

    I have also asked them ifsemi-quantative results can be obtainedor whether only a Yes or No answer can be obtained. I will let you know as well, what Ifind out.
