Human G-CSF ELISA Kit (ab100524)

We have not tested the GCSF ELISA kit with Cynomolgus monkey serum. However, when looking at the human GCSF sequence our kit recognizes and testing the cross-reactivity to the Cynomolgus sequence, there are a number of predicted matches, including granulocyte colony-stimulating factor (isoforms X1-4), with which it has 94% homology. Thus, it is likely that the kit will work with Cynomologus serum. Especially since we have a 90 day 100% risk-free policy for ELISAs, there is no risk in this case to try it.

Please keep in mind though that there are a number of other predicted matches (other than GCSF) with varying homology that it may cross-react with and there is not a 100% sequence match. So you will have to consider this when interpreting any results.

The standard recombinant protein of ab100524 was expressed in E.coli.

The standard we use for this kit (ab100524) is recombinant human G-CSF: 18.7 kDa protein; 174 amino acid residues. We have not tested this kit with a met-GCSF, but the following amino acid sequence of the recombinant standard used should help the customer determine whether or not the antibody pairs used will recognize the met-GCSF as well:

Serum can be stored at -80C for a month, this will be having minimal affect on the assay.

The strips come with a rack where the single strips can be inserted.

This rack will fit all standard plate readers.

I am sorry, I cannot check if this is also true for your plate reader. Please ask the supplier of the instrument.

I can confirm that with ab100524 it is not necessary to use the whole plate at one time.

The plate comes in convenient 12 strips x 8 wells.

Please follow the instruction of the protocol when storing opened plate and solutions:

"

ab100524 may be stored for up to 6 months at 2 to 8°C from the

date of shipment. Standard (recombinant protein) should be stored

at -20°C or -80°C (recommended at –80°C) after reconstitution.

Opened Microplate Wells or reagents may be store for up to 1 month

at 2 to 8°C. Return unused wells to the pouch containing desiccant

pack, reseal along entire edge. Note: the kit can be used within one

year if the whole kit is stored at -20°C. Avoid repeated freeze-thaw

cycles."

We still have not tested whether the kit can be used to quantitate PEGfilgrastim, therefore the original response, "to spike in pegylated G-CSF and see" is still the only option for answering this question.

Thank you for your inquiry.  That is a good question.  I reviewed our testing records on ab100524 but unfortunately I was not able to find any data about pegylated G-CSF.  Therefore we don’t know if this kit will also detect the pegylated form.  The only way to know would be to spike in pegylated G-CSF and see.   I wish I could be of more help, but please let us know if you have any other questions.
