    Abcam’s Apolipoprotein B (APOB) Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Apolipoprotein B concentrations in Human plasma, serum, CSF and cell culture samples.


    An Apolipoprotein B specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently an Apolipoprotein B specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Complex is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of Apolipoprotein B captured in plate.

Only the assays for APOB (ab108807) and APOE (ab108813) have been tested with CSF. They are suitable for this kind of sample. The APOA1 assays have not been tested with CSF yet, but there are plans to test ab108803. Kit ab108804, the competitive ELISA for APOA1, will not be tested with CSF, and it is not known if it is suitable for assays of CSF.

The lyophilized powder standard is supplied at 0.75 ug. Reconstitute the 0.75 ug of Apo B Standard with 3 ml of EIA Diluent to generate a solution of 0.25 ug/ml. Any leftover solution should be stored at -20C.
The supplying lab may be able to provide us with an additional standard for you to purchase. If you are interested in this, please let me know and I will ask our Products Team if they can add this to the catalogue for you.

I believe this kit includes 0.75ug of standard. Based on the protocol, this provides you 3 mls of 0.25ug/ml of the highest concentration of standard. This starting concentration is subsequently diluted in 2x serial dilutions to produce the curve. Since only 50ul are required per standard well, you should have plenty of standard volume to test the assay kit.
