On this picture, cells were infected for 9 hours with HSV KOS. I could detect infected cells as soon as 3 hours post-infection. I could also detect cells infected with 27LacZ virus (ICP27 mutant). Extremely bright and clean staining, no background on uninfected control cells, could probably dilute it much further!
Fixation is 10 minutes with 1.9 % formaldehyde in 2 % sucrose, permeabilisation is 0.5 NP40 in 10 % sucrose for 10 minutes.
