sELISA: Use at an assay dependent dilution. Can be paired for ELISA with Mouse monoclonal [C5] to C Reactive Protein (ab8279) and Mouse monoclonal [C7] to C Reactive Protein (ab8280).



Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

    I am planning to test a C-reactive protein sandwich ELISA assay and was hoping that you could suggest the most suitable antibody pair for this application from your range?


    Also, on your website it lists the molecular weight of the C-Reactive Protein subunit as being 23 kDa, but also states that the protein forms a pentamer in solution. Please can you advise on whether in a solution ELISA assay I would be detecting the subunits alone (23 kDa) or the pentameric form (115000 kDa)?

    I have confirmed with the lab and most of our C Reactive Protein antibodies, except C3 and C4, react with both monomer and pentamer form of CRP. (As our clones C3 and C4 do not recognize CRP in the absence of Ca2+ ions, we suggest that they could not recognize CRP monomer, but we have never experimentally tested this.)

    Thank you for your response. I have looked into the biotinylation kits you have suggested. I would like to see if the following combination will work for a CRP sanwhich ELISA:

    -streptavidin coated surface-
    Primary Antibody - ab8280 Anti-CRP antibody modified with ab102863 EasyLink Biotin.
    Antigen - ab51784 CRP (Human)
    Secondary Antibody - ab19174 Anti-CRP antibody (FITC) or ab24462 Anti-CRP (HRP)

    My only concern is whether the Primary antibody will compete with the Secondary antibody for antigen binding? Will this be an issue?

    The combination of biotinylated ab8280 and HRP-conjugated ab24462 should work, as these two clones have been tested together and are known to be an effective pair for sandwich ELISA. The goat polyclonal ab19174 has not been tested with ab8280. It may work but I think you are better off with ab8280 combined with ab24462. In addition, the FITC conjugate will require a fluorescent plate reader, in case you were not already aware of that.

    I have seen you do have anti-CRP HRP conjugate, but do you have any anti-human crp biotin conjugated antibody? I have seen you have anti-CRP (biotin) anti-rat ab86246, but I would need anti-human.

    Thank you for contacting. My search also shows just the single biotinylated antibody, anti-rat CRP, ab86246.

    We do have biotinylation kits which are very easy to use. For instance, the rabbit polyclonal antibody ab31156 can be biotinylated with ab102863 or ab102866, depending on the intended use of the antibody. (Please see the Product Overview section of each datasheet for an explanation of the differences between the two products).

    We are currently looking for a good pairing for C-Reactive protein antibodies and wonder if you guys could recommend any pair of antibodies? The antibodies will be used in sandwich ELISA and we are looking for antibodies which are already paired by Abcam for optimal binding and lowest cross reactivity of capture and detection antibody.
    We've noticed that it is recommended to pair antibodies according to their C number, for example C2-C6, C5-C6 and C7-C6, information found on website: https://www.abcam.com/C-Reactive-Protein-antibody-C6-ab8278.html.
    It is best to pair antibodies to their C numbers as mentioned?

    We do not use the terminology C number. These does not depicts any information other the clone numbers for a particular antibody.
    The sELISA pairs we recommend are
    The best pair is C2-C6 (ab24461 detection - ab8278 capture)
    C5-C6 (ab8279 capture - ab24462 detection)
    C7-C6 (ab8280 capture - ab24462 detection)
    Please ab24462 is a HRP conjugated anti C reactive protein antibody; due to the conjugation it will be a good detection antibody.
