    This protein is a component of the heterogeneous nuclear ribonucleoprotein (hnRNP) complexes which provide the substrate for the processing events that pre-mRNAs undergo before becoming functional, translatable mRNAs in the cytoplasm. Mediates pre-mRNA alternative splicing regulation. Inhibits, together with CUGBP1, insulin receptor (IR) pre-mRNA exon 11 inclusion in myoblast. Binds to the IR RNA. Binds poly(RG).

Your antibody ab10689 is supposed to recognize hnrpH and hnrpF. There are several hnrpH's - H1, H2 and H3. H1 and H2 are very highly homologous, while H3 is not, so presumably the ab doesn't recognize H3. However, does it recognize both H1 and H2, which are slightly different m.w.'s (though distinguishable on SDS-PAGE), in addition to hnrnpF? Also, your other anti-hnrpH antibody, ab10374 - does it recognize H1, H2 and/or F? (the website states it doesn't recognize H3)

Thank you for your enquiry. The epitope recognized by ab10374 has been mapped to a region between residue 400 and the C-terminus (residue 448) of human Heterogeneous Nuclear Ribonucleoprotein H1 using the numbering given in entry NP_005511.1 (GeneID 3187, accession number P31943). Although testing for reactivity with H2 has not been done, alignment of this H1 region with the human H2 protein (accession number P55795) shows 96% identity, so we predict the antibody will react with H2. The file for this alignment can be found at this link: http://www.ebi.ac.uk/cgi-bin/clustalw/result?tool=clustalw&jobid=clustalw-20070719-16083294&poll=yes Alignment with the human H3 protein (P31942) showed 24% identity. Question

I am scientific from [] in Spain. Recently we have bought an antibody from you. The order number is: 116A/3848/05. We have several problems with this antibody as you can see in the file I enclose to you as an attachment. I bought from you the antibody anti hnRNP H (ab10374). I have had problems to see the protein by WB. I looked the recommended dilution in the data sheet of the product and I think that there is a mistake, since you say that for WB it is recommended to use a concentration of 10000-30000 microgram per milliliter. The stock concentration is 1mg/ml, so it’s impossible to prepare this dilution. You have a link in the web that shows different concentrations, so I used the antibody at the following concentrations: - 0,004 mcg/ml - 0,05 mcg/ml - 1 mcg/ml I couldn’t see the hnRNP H in any case. I used this concentrations in different types of cells: 293T (human), Hela (human), COS (monkey), NLB2 (dog), BHK21 (hamster), and H5 (insect). For each type of cell I used 20 micrograms of total protein. This is the WB: the first one is 0,05mcg/ml, and the second one is 1mcg/ml. I would apreciate receiving a new antibody free of cost because the one we got is not working properly. Beside that, I submit you different images of the other antibodies we bought since you give 100 points per each one. Thank you very much,

I'm sorry to hear you are experiencing a problem with ab10374 in WB. You are absolutely correct, there was indeed an error on the datasheet and it has been corrected to " use at a dilution of 1:10 000 to 1:30 000, therefore this would be equivalent to 0.03ug/ml-1ug/ml, therefore the range of dilutions you have tested is very adequate. This antibody has not been tested in species other than human so it may be that part of the problem you are experiencing is due to a lack of cross reactivity with monkey, dog and hamster however in our hands HeLa nuclear lysate gave a very strong signal. As you do not specify if your samples were nuclear lysates I suspect you do not do a nuclear extraction to concentrate the protein and this may be the reason for the lack of signal you currently are experiencing. The multiple faint bands I see on your image you kindly provided can be due to a number of reasons and without a detailed protocol it is difficult to give you some suggestions.
