    This antibody can be applied for staining of monocytes and up to 10 % of lymphocytes from peripheral blood of healthy donors. Blast cells of patients with AMMonL (M5 following FAB classification), AMMonL (M4) and hairy cells leukemia were ab24093 positive. The antigen, defined with this mAb, is particularly expressed on blood cells from patients with infectious mononucleosis and CLL. Histiocytes and macrophages were also positive. Malignant cells from patients with AML (M1 and M2), T-ALL, B-ALL were ab24093 negative.

    HL-60 cells are used as an in vitro model of acute promyelocytic leukaemia and for differentiation and apoptosis studies. The HL60 cell line was established in 1977 from a patient with acute myeloid leukaemia. The cells largely resemble promyelocytes but can be induced to differentiate terminally in vitro. Some reagents cause HL60 cells to differentiate to granulocyte-like cells, others to monocyte/macrophage-like cells. The HL60 cell genome contains an amplified c-myc proto-oncogene; c-myc mRNA levels are correspondingly high in undifferentiated cells but decline rapidly following induction of differentiation.

Question

We are searching for an antibody that is only specific for undifferentiated HL-60 cells. Do you think that IPO-M6 can be used for immunocytochemistry, and has it ever been tested for ICC by abCAM? Do you have any other antibodies that you believe could work? Lastly, since you don’t have record of a publication using IPO-M6, could you provide us with a small aliquot to test?

Thank you for contacting us. The antibody ab24093 has not yet been tested in ICC and do not know how well it might work. But I think there would be a good chance of it being successful as it works well in frozen sections (similar protocol) and flow cytometry (similar sample preparation). In general, we are not able to offer small aliquots of any of our products, but have our guarantee (https://www.abcam.com/abpromise) for tested species/applications and the discount program for untested species/applications.
