    Inquiry: Sample: 40 ug of total protein from control or HERC6 shRNA knockdown HeLa cells (RIPA lysates) were loaded on 7% SDS-PAGE blocking: 5% non-fat milk detection: Femto reagent (Pierce) primary antibody dilution 1:1000 in TBST+ 0.3% BSA + 0.02% NaN3, over night at 4 degree secondary antibody 1:1000 in TBST, 1h at RT wash steps always in-between 3x 10 minutes at RT in TBST Results: antibody detected over-expressed HERC6 in HEK293T cells without any problem, but there was no specific endogenous HERC6 band in HeLa
