    ab19346 has been successfully used in Western blot procedures. By Western blot, this antibody detects bands at ~100 kDa and ~115 kDa representing the non-glycosylated and glycosylated form of HCN2 in rat brain extract.

    Immunofluorescent analysis of Neuro-2a (Mouse neuroblastoma cell line) cells on 70% confluent log phase labeling HCN2. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labelled with ab19346 at 2 µg/mL in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with Goat anti-Rabbit IgG (H+L) secondary antibody, Alexa Fluor® 488 conjugate at 1/2000 dilution for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with DAPI. F-actin (Panel c: red) was stained with Alexa Fluor® 555 Rhodamine Phalloidin (1/300 dilution). Panel d is a merged image showing localization. Panel e is a no primary antibody control. The images were captured at 60X magnification.

Can you tell me where I can purchase the immunising peptide for this antibody and what their Western blot protocol is including the preparation of lysates?

Answer

Thank you for your enquiry. Unfortunately we do not sell the immunising peptide for this antibody. The Western blot protocol used to test this antibody can be found under our Protocol tab. Regarding lysate prep, you should be able to use a standard lysis buffer (75 mM Tris-HCl, pH 7.2, 200 mM NaCl, 1% Tween 100, 100 mM EDTA, 1 mM DTT with protease inhibitors). Please note, HCN2 has an N-linked glycosylation in the brain. This antibody detects a band at ~115 kDa (due to glycosylation). However this band can be reduced to ~100 kDa by the addition of PNGase F.
