    Question

    I used HCC1 antibody (ab12326) and my questions are: 1. The predicted MW is 63 kDa, but why in your product sheet shown that was about more than 70kDa? Which one is correct? 2. Any suggestion to reduce the background of this antibody?- it seems a litter bit high for my experiment. Thanks

    Thank you for your enquiry. I am sorry for the confusion, HCC1 antibody (ab12326) detects a protein with a predicted molecular weight of 58KDa. However, the actual band as detailed on the western image is more like 70KDa. The observed molecular weight of a protein can differ from its predicted molecular weight for a number of systematic biological reasons. The mass of a protein can be increased by post-translational modifications, such as glycosylation, ubiquitination, and sumoylation, among others, while the mass can be decreased by alternative splicing and endoproteolytic cleavage. Additionally, there are reports of altered migration for some subsets of proteins, including highly acidic, highly basic, and arginine-rich proteins. I would like to recommend that you try performing an overnight incubation at 4oC using this antibody. Additionally incorporating 3% BSA as a blocking agent as an alternative to non-fat milk often leads to a reduction in background noise.     I had a couple of questions regarding the anti-HCC1 ab9845 rabbit polyclonal antibody. I have been using the ab12326 polyclonal antibody for my western blotting of HCC1 protein, but have had no success with co-/immunoprecipitation using it, and so wanted to try the other anti-HCC1 antibodies. From the web site datasheet I understand that the ab9845 is raised against purified recombinant hHCC-1. I would like to know if anything is known further as to which epitope(s) is recognised by this antibody, for eg, whether the epitope is N-or C-terminal, etc. Also I would appreciate it if you could share any information relating whether the ab9845 antibody has been used, successfully or otherwise, in immunoprecipitation and ChIP assays. Any information, like cleanliness of detection, dilution or other conditions, would be very helpful.

    Thank you for your enquiry. First, I'm pleased to hear that ab12326 is working out for you in Wetern blotting. Ab12326 has not yet been tested for application in IP. I do suggest that you submit a review regarding your experiences with the antibody - in return we will award you 50 points with the Abcam Loyalty Scheme which can be redeemed on a number of rewards. As for ab9845, it is a polyclonal antibody and will therefore consist of a mixed population of IgGs each of which will recognise a different epitope.
