    This blot was produced using a 4-12% Bis-tris gel under the MES buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour before being incubated with ab16502 overnight at 4°C. Antibody binding was detected using the Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed (ab216773) at a 1:10,000 dilution for 1hr at room temperature and then imaged using the Licor Odyssey CLx.

    Merged signal (red and green). Green - ab16502 observed at 70 kDa. Red - loading control, ab8245, observed at 37 kDa Goat anti-Mouse IgG H&L (IRDye® 680RD) ab216776.

  • All lanes : Anti-Lamin B1 antibody - Nuclear Envelope Marker (ab16048) at 1 µg/ml (Anti-Lamin B1 antibody - Nuclear Envelope Marker)

    Lane 1 : Wild-type HAP1 nuclear lysate, 10 ug
    Lane 2 : Lamin B1 knockout HAP1 nuclear lysate

Application
In-Cell ELISA
We tested this product for in cell ELISA for antibody validation, and found it worked well.
We used 6 different rabbit antibodies against the same target and performed in cell ELISA with wild type Hela cells and two different CRISPR mediated knockout cells as positive and negative controls, respectively. After staining with primary antibodies, cells were stained with x10,000 diluted this product and analyzed by using Odyssey CLx (LI-COR).
