  • IHC image of beta Actin staining in normal human colon, formalin-fixed and paraffin-embedded tissue*. The section was pre-treated using pressure cooker heat mediated antigen retrieval with sodium citrate buffer (pH6) for 30mins. The section was incubated with ab8227, 3µg/ml overnight at +4°C. An HRP-conjugated secondary (Ab97080, 1/2000 dilution) was used for 1hr at room temperature. The section was counterstained with haematoxylin and mounted with DPX.

    The inset negative control image is secondary-only at 1/500 dilution.

Western blot
This item was applied as a secondary Ab for my WB. 1:10,000 dilution was applied for chemiluminescence reaction afterwards. I also conducted the procedure with secondary Ab probing only (right side of the blot) and the result showed no non-specific binding for this Ab. I am satisfied for this item in my WB assay.

Immunohistochemistry (Frozen sections)
Sections were thawed and fixed in cold acetone. Endogenous peroxidase and serum blocking (10% normal horse serum) were performed. Primary antibody was incubated with ab9722, at 5µg/ml overnight at 4°C. An HRP-conjugated secondary (ab97080 at 1/300 dilution) was used for 30min at room temperature. Sections were counterstained with haematoxylin and mounted with DPX.
A negative control was included using an isotype IgG at the same concentration as the primary antibody.

I am sorry I am unable to get the specific protocol for chicken tissues. My colleague from laboratory has suggested that any general IHC-P protocol would work and the staining can be improved with optimization.

While I did not receive the image which you have referenced I would like to offer so help regarding your questions. Generally specific details regarding secondary antibody incubation and concentration as will need to be determined empirically in the lab. These may vary considerable depending upon the secondary antibody used. This is the same for exposure times. We do offer a number of guidelines for information on our datasheet however. Please remember that these are suggested starting points and you will likely need to adjust these to find the optimal ranges.

The secondary antibody AB97080 Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (HRP), pre-adsorbed (ab97080) was used at 1/5000 dilution. In this experiment ECL detection was used with a 20 minute exposure time. This product may be found at this location:

The secondary antibody AB65484 Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (HRP), pre-adsorbed (ab97080) was used at 1/5000 dilution. In this experiment ECL detection was used with a 150 second exposure time. Unfortunately this product is no longer available.

A third experiment is shown on the datasheet; however we do not have specific information as to the product number of the secondary antibody used.

Abcam takes product quality seriously and we recently undertook an in-depth study examining storage conditions and antibody performance. After over 13,000 individual ELISA experiments, we have determined that storing our antibodies at various temperatures of up to 45°C for 1 week does not impact their activity. I would be happy to share a summary of our findings with you; please let me know if you would like to see this.

As a precautionary measure only, we ship our antibodies in packaging with ice packs to provide extra temperature stability in transit. Once you have received any products, we recommend toplease follow the long-term storage instructions on the datasheet as soon as possible. This will help to ensure that the product remains stable and that we can continue to provide our guarantee.

In this case, considering our findings described above, I would suggest that the antibody should not be significantly affected from being at room temperature for 3 days. We recently redesigned our packing. A decision was taken to label these with 'Temperature sensitive - store according to datasheet instructions' in order to cover all the different products we ship at different temperatures.This instruction should prompt the recipient to check the datasheet, or check with the end user, as soon as possible.
