Hi
I am looking to order gold labelled secondaries- eg Goat Anti-Mouse IgG+IgM H&L (18nm Gold) preadsorbed (ab105290) for labelling proteins which I will then determine the concentration of via mass spec by measuring gold concentration.
I need to know the concentration of the antibodies so I can work out the concentration of gold. Can you tell me what concentration they are supplied at?

We do not determine the antibody concentration for this gold conjugated antibody because during preparation monomers from multimers are separated on density gradients. We do not have any reliable way to measure the antibody concentration on the monomers because the preparation is stabilized with albumin before and after centrifugation.

The colloidal gold-antibody complex is formed by passive adsorption. We express the concentration of the complex in terms of adjusted OD at 520 nm; the complex is tested for antigen binding and the reported OD at 520nm reflects those gold particles to which active antibody is bound. For our gold conjugates, the OD at 520 nm is 2.0. We do not have a value for the molar ratio of gold particles to antibody.
