Inquiry: Good morning- I am inquiring about an intracellular staining protocol for the anti-gp100 antibody (ab117853) with the FITC-conjugated anti-mouse IgG2b (ab98702). There was a reference made to some staining protocols but I could not locate the actual protocols. Please see below for a portion of a post that I found on the website. 'For gp100, we have unconjugated antibodies but neither is in a formulation that is compatible with the conjugation kits for FITC. What I recommend is using the unconjugated anti-gp100, ab117853, followed by FITC-conjugated anti-mouse IgG2b, ab98702, pre-adsorbed to remove reactivity with mouse IgG1 (which is the isotype of ab52460, anti-cytokeratin).' Thank you for your inquiry and your interest in our antibodies. The recommendation you found on the website, was specific in regards to make a double staining with another specific antibody.

I can recommend you to follow the classical intracellular staining protocol for using ab117853 as per the link here. The antibody should work perfectly well in these conditions. Optimal dilution should be determined by you in the laboratory, as a starting point I would recommend dilutions of 1/50, 1/100 and 1/200 as we had good results with 1/100 dilution.

The secondary antibody can be used in a first test at 1/50 to be sure to be saturating. When you have determined the optimal dilution of the primary antibody, I can recommend to test whether the secondary antibody can also be further diluted.

We have a PE- conjugated antibody against cytokeratin, catalogue item ab52460:

For collagen I, we have only one unconjugated antibody that has been tested in flow cytometry. We do, however, have a kit that enables one to easily conjugate the antibody to PE.

For gp100, we have unconjugated antibodies but neither is in a formulation that is compatible with the conjugation kits for FITC. What I recommend is using the unconjugated anti-gp100, ab117853, followed by FITC-conjugated anti-mouse IgG2b, ab98702, pre-adsorbed to remove reactivity with mouse IgG1 (which is the isotype of ab52460, anti-cytokeratin). This should allow you to apply both antibodies to the same sample without cross-reaction of the secondary anti-IgG2b with mouse IgG1.
